摘要:目的观察百合多糖联合BMSCs移植是否影响小鼠肺纤维化的形成。方法培养小鼠BMSCs做移植备用。90只SPF小鼠按随机数字表法分为对照组(C)、模型组(M)、百合多糖联合BMSCs组(A)、BMSCs组(B)、百合多糖组(L),每组18只。建立肺纤维化小鼠模型,24 h后各组给予相应处理。第7、14、28天行肺组织病理评分。第28天观察各组肺系数、肺组织胶原沉积、羟脯氨酸(HYP)含量、Western blot检测肺组织TNF-α和NF-κB蛋白表达情况。结果与C组相比,M组肺系数、病理形态评分、HYP含量、肺组织TNF-α、NF-κB表达均增高(P<0.05)。与M组相比,A组、L组各指标均降低(P<0.05),B组第28天病理评分降低(P<0.05)。M组有大量胶原沉积,各干预组胶原沉积均较轻。结论百合多糖联合BMSCs移植后抑制肺纤维化小鼠肺组织的TNF-α、NF-κB的表达,减轻肺纤维化的病理损伤,联合干预的效果强于单用百合多糖或单用BMSCs移植。
Objective To determine the effect of lily polysaccharides combined with bone marrow mesenchymal stem cells (BMSCs) transplantation on bleomycin-induced pulmonary fibrosis mice. Methods BMSCs of mice were cultured in vitro. Ninety healthy SPF mice were randomly divided into a control group (C), a model group (M), a bleomycin + lily polysaccharides combined with BMSCs transplantation group (A), a bleomycin + BMSCs transplantation group (B), and a bleomycin + lily polysaccharides group (L). The mice of each group were treated with the corresponding measures at 24 h after the mouse models of pulmonary fibrosis were constructed by bleomycin. On the 7th, 14th and 28th day, the mice were sacrificed and the pathological scores of lung tissues were determined by HE staining. On the 28th day, the lung coefficient (LC) and hydroxyproline (HYP) content were determined, lung collagen deposition was observed by Masson trichrome staining, and the expression levels of TNF-α and NF-κB in lung tissues was measured by Western blot analysis. Results There were significant differences in the LC, HYP content, pathological score, and the expression levels of TNF-α and NF-κB in lung tissues between group C and group M (P〈0.05). Compared with group M, those indices significantly decreased in group A and group L (P〈0.05), and the pathological score on the 28th day significantly decreased in group B (P〈0.05). The collagen deposition in group M was greater than that in group A, group L and group B. Conclusion Lily polysaccharides combined with BMSCs transplantation can reduce the expression levels of TNF-α and NF-κB in lung tissues and pulmonary pathological changes in bleomycin-induced pulmonary fibrosis mice. The effect of combined intervention is better than that of lily polysaccharides or BMSCs transplantation alone.
