摘要:目的:观察黄芪汤对^(12)C^(6+)辐射脑损伤模型鼠脾脏细胞因子、外周血象及Survivin蛋白表达的影响,以揭示其防辐射分子机制。方法:50只SPF级Wistar大鼠随机分为正常对照组,单纯辐射模型组,黄芪汤高、中、低剂量组,每组10只。黄芪汤高、中、低剂量组分别给予黄芪汤18.0、9.0、4.5g·kg^(-1)·d^(-1),灌胃2周。干预7d后除正常对照组外,其余各组脑组织给予4Gy ^(12)C^(6+)离子束单次照射,辐射后7d处死各组大鼠。HE染色观察大鼠肾组织的病理形态变化,ELISA测定大鼠脾脏IL-1β、IL-2和IL-6的含量,血细胞分析仪检测大鼠外周血细胞的数量变化,RT-PCR及Western Blot测定大鼠肾组织Survivin的mRNA及蛋白表达。结果:与正常对照组比较,单纯辐射模型组体质量、胸腺指数与脾脏指数、脾脏IL-2含量、外周血象WBC和RBC数量和肾组织Survivin蛋白表达均显著降低,而脾脏IL-1β和IL-6的含量均升高(P<0.05,P<0.01);单纯辐射模型组肾小球系膜细胞明显增生,肾小管间质血管明显扩张充血,肾小管管腔狭窄、不规则。与单纯辐射模型组比较,黄芪汤高剂量组体质量、脾脏IL-2含量和PLT数量均显著升高,脾脏IL-1β和IL-6含量均下降,黄芪汤低、中剂量组PLT数量则显著减少,黄芪汤中、高剂量组肾组织Survivin基因表达和蛋白表达、外周血WBC和RBC数量均显著升高,黄芪汤各干预组胸腺指数和脾脏指数均升高(P<0.05,P<0.01);黄芪汤高剂量组可见肾小球系膜细胞增生情况显著改善,肾小管轮廓清晰。结论:黄芪汤对^(12)C^(6+)辐射脑大鼠诱发的旁效应脏器损伤具有一定的保护作用,其作用机制可能是通过增强机体的免疫力、抑制肾组织的细胞凋亡和调节外周造血系统,促进体质量恢复来达到的。
Objective:To observe the effects of Huangqi Decoction on cell factor,peripheral blood and Survivin protein expression in rats with ^(12)C^(6+)radiation brain damage,and reveal its molecular mechanism of radiation protection.Methods:Fifty Wistar rats were randomly divided into five groups:normal control group,radiation alone model group,highdose,middle-dose and low-dose Huangqi Decoction group.After 7 days of routine feeding,the high-dose,middle-dose and low-dose Huangqi Decoction group were given Huangqi Decoction(18.0,9.0,4.5 g·kg^(-1)·d^(-1))by gavage for one weeks in advance.The normal control group and the radiation alone groups were given the equal normal saline.After gavage for 7 d,rats of all groups except the control group were radiated by 4 Gy ^(12)C^(6+)ion once.After radiation for 7 d,all groups of rats were executed.The pathomorphism changes of renal tissue were observed by HE,the contens of IL-1β,IL-2 and IL-6 in spleen tissue were checked by ELISA,the number changes of peripheral blood cells were detected by hematology analyzer,the amount of mRNA and protein expression of Survivin in kidney was examined with RT-PCR and Western Blot.Results:Compared with the normal control group,the body weight and the indexes of thymus and spleen significantly decreased,the IL-2 level in spleen significantly reduced,the number of WBC and RBC in peripheral blood significantly cut down,the gene and protein of Survivin in kidney tissue significantly decreased,whereas the levels of IL-1βand IL-6 in spleen significantly raised(P<0.05,P<0.01).The mesangial cells proliferated obviously,interstitial vessels of renal tubules dilated and congested obviously,the lumen of renal tubules was narrow and irregular in the radiation alone model group.Compared with the radiation alone model group,the body weight,IL-2 level in spleen and the number of PLT in peripheral blood obviously raised in high-dose huangqi decoction group,yet the levels of IL-1βand IL-6 in spleen significantly decreased in high-dose Huangqi Decoction group,the PLT number obviously decreased in low-dose,middle-dose Huangqi Decoction group,the gene and protein of Survivin in kidney tissue,the number of WBC and RBC in peripheral blood significantly increased in middle-dose,high-dose Huangqi Decoction group,the indexes of thymus and spleen obviously raised in Huangqi Decoction intervention group(P<0.05,P<0.01).The proliferation of mesangial cells was improved and the contour of renal tubules was clear in high-dose Huangqi Decoction group.Conclusion:Huangqi Decoction has a certain protective effect on the organ damage caused by ^(12)C^(6+)radiation brain induced bystander effect in vivo.Huangqi Decoction reduced the organ injury in rats with ^(12)C^(6+)radiation brain through immunity enhancement,inhibition of apoptosis in renal tissue and the hematopoietic system rebuilt and body weight regain.
