摘要:目的观察黄芪乌梅配方颗粒对GK大鼠血糖及胰岛素敏感性的影响。方法将32只雄性GK大鼠随机分为模型组、二甲双胍组、黄芪乌梅1.3 g/mL组、黄芪乌梅5.2 g/mL组,每组8只。另选择8只同周龄、同种系雄性Wistar大鼠作为正常组。模型组及正常组给予生理盐水灌胃,二甲双胍组给予盐酸二甲双胍肠溶片0.15 g/mL灌胃,黄芪乌梅1.3 g/mL组和黄芪乌梅5.2 g/mL组分别给予相应剂量黄芪乌梅液灌胃,均1次/d,连续10周。各组大鼠分别于灌胃前及灌胃2,4,6,8,10周进行口服葡萄糖耐量试验(OGTT);末次灌胃结束后,测定各组大鼠血清空腹胰岛素(FINS)、肿瘤坏死因子-α(TNF-α)、游离脂肪酸(FFA)水平,计算胰岛素敏感性指数(HOMA-ISI)、胰岛素抵抗指数(HOMA-IR)、胰岛β细胞功能指数。结果灌胃2,4,6,8,10周,二甲双胍组大鼠空腹血糖水平和血糖曲线下面积均明显低于模型组(P均<0.05);灌胃2周,黄芪乌梅1.3 g/mL组和黄芪乌梅5.2 g/mL组大鼠仅空腹血糖水平明显低于模型组(P均<0.05);灌胃4周,黄芪乌梅5.2 g/mL组大鼠血糖曲线下面积明显低于模型组(P<0.05);灌胃6周和10周,黄芪乌梅1.3 g/mL组和黄芪乌梅5.2 g/mL组大鼠空腹血糖水平和黄芪乌梅5.2 g/mL组大鼠血糖曲线下面积均明显低于模型组(P均<0.05);灌胃8周,黄芪乌梅1.3 g/mL组和黄芪乌梅5.2 g/mL组大鼠血糖曲线下面积和黄芪乌梅5.2 g/mL组大鼠空腹血糖水平均明显低于模型组(P均<0.05)。灌胃结束后,模型组大鼠FINS、HOMA-IR、TNF-α、FFA水平均明显高于正常组(P均<0.05),HOMA-ISI和HOMA-β均明显低于正常组(P均<0.05);二甲双胍组和黄芪乌梅5.2 g/mL组大鼠FINS、HOMA-IR、TNF-α、FFA水平均明显低于模型组(P均<0.05),HOMA-ISI和HOMA-β均明显高于模型组(P均<0.05);黄芪乌梅1.3 g/mL组HOMA-IR、TNF-α、FFA水平均明显低于模型组(P均<0.05),HOMA-β明显高于模型组(P均<0.05)。结论黄芪乌梅配方颗粒能明显降低GK大鼠血糖,其可能通过降低血清TNF-α和FFA水平而改善GK大鼠胰岛素敏感性和胰岛β细胞功能。
Objective It is to observe the effect of Huangqi Wumei formula granules(HWFG) on blood glucose and insulin sensitivity in GK rats. Methods Thirty-two male GK rats were randomly divided into model group, metformin group, HWFG 1.3 g/mL group, HWFG 5.2 g/mL group, 8 rats in each group. Another 8 male Wistar rats of the same age and the same lineage were selected as the normal group. The rats in the model group and the normal group were given normal saline by gavage, the ones in the metformin group was given metformin hydrochloride enteric-coated tablets 0.15 g/mL by gavage, and the ones in the HWFG 1.3 g/mL group and HWFG 5.2 g/mL group were administered with corresponding doses of HWFG solution by gavage, all 1 time/d for 10 consecutive weeks. Oral glucose tolerance test(OGTT) was performed in the rats of each group before and at 2, 4, 6, 8, and 10 weeks of treatment. After the last administration, the levels of serum fasting insulin(FINS) and tumor necrosis factor-α(TNF-α) and free fatty acid(FFA) were determined, the insulin sensitivity index(HOMA-ISI), insulin resistance index(HOMA-IR), and islet β-cell function index were calculated. Results After 2, 4, 6, 8, 10 weeks of gavage, the fasting blood glucose level and the area under the blood glucose curve of the metformin group were significantly lower than those of the model group(P<0.05). After 2 weeks of gavage, only the fasting blood glucose level of the rats in the HWFG 1.3 g/mL group and HWFG 5.2 g/mL group was significantly lower than that in the model group(P<0.05). After 4 weeks of gavage, the area under the blood glucose curve of the HWFG 5.2 g/mL group was significantly lower than that in the model group(P<0.05). After 6, 10 weeks of gavage, the fasting blood glucose levels of rats in the HWFG 1.3 g/mL group and the area under the blood glucose curve in the HWFG 5.2 g/mL group were all significantly lower than those in the model group(all P<0.05). After 8 weeks of gavage, the area under the blood glucose curve of the HWFG 1.3 g/mL group and the 5.2 g/mL HWFG group and the fasting blood level glucose in the HWFG 5.2 g/mL were significantly lower than those in the model group(P<0.05). At the end of the treatment, the levels of FINS, HOMA-IR, TNF-α, and FFA in the model group were significantly higher(P<0.05), and HOMA-ISI and HOMA-β were significantly lower than those in the normal group(P<0.05);the levels of FINS, HOMA-IR, TNF-α and FFA in the metformin group and the HWFG 5.2 g/mL group were significantly lower(P<0.05), and HOMA-ISI and HOMA-β were significantly higher than those in the model group(P<0.05);the levels of HOMA-IR, TNF-α, FFA in the HWFG 1.3 g/mL group were significantly lower(P<0.05), and HOMA-β was significantly higher than those in the model group(P<0.05). Conclusion Huangqi Wumei formula granules can significantly reduce blood glucose in GK rats, which may improve insulin sensitivity and islet β-cell function in GK rats by reducing serum TNF-α and FFA levels.
