摘要:目的:研究稳心草黄酮类化合物对大鼠肺动脉平滑肌细胞(PASMs)增殖和TGF-β表达水平的影响,探讨其防治肺动脉高压的疗效及作用机制。方法:组织帖块法培养大鼠肺动脉平滑肌细胞,MTT法确定高、中、低剂量分组及作用时间。MTT法进行细胞计数,流式细胞仪进行细胞周期测定,ELISA法进行细胞培养夜TGF-β含量测定。结果:贴块培养的细胞经电镜鉴定为动脉平滑肌细胞;台盼蓝染色倒置显微镜观察表明细胞生长状态良好。黄酮类化合物高、中剂量组细胞计数明显减少、细胞周期G0/G1期细胞增多,S期、G2/M期细胞减少;细胞TGF-β含量明显降低,与空白对照组比较有统计学意义(P<0.05);低剂量组与空白对照组比较无统计学意义(P>0.05)。结论:稳心草黄酮类化合物可抑制大鼠肺动脉平滑肌细胞增殖,延缓细胞周期进程,调节细胞分泌TGF-β水平。
Objective:To explore the mechanism of hypericum attenuatum chois flavonoid on the prevention of pulmonary hypertension by observing the effects on rats' pulmonary arterial smooth muscle cells(PASMCs) proliferation and transforming growth factor beta(TGF-β).Methods:PASMCs were attachment-block cultured and certified with invert light microscope and electron microscope,dosages(low,middle,high) and time were determined by MTT.The number of cells was determined by MTT method,cycle of cells was examined by flow cytometer,TGF-β content in tissue cultured fluid was determined by ELISA method.Results:The attachment-block cultured cells were certified with electron microscope to be PASMCs,the number of cells in high and middle dose hypericum attenuatum chois flavonoid groups was decreased obviously,the rate at G0/G1 stage of cells cycle was increased significantly and the rate at S and G2/M stages of cells cycle was decreased obviously in high and middle dose groups,the TGF-β contents of middle and high dose groups were reduced significantly to compare with that of the control.Conclusion:Hypericum attenuatum chois flavonoid could inhibit the proliferation of PASMCs,prolong the progress of cells cycle,regulate the TGF-β level.
