摘要:目的观察生脉硒对异丙肾上腺素诱导大鼠心肌缺血损伤的影响,探讨其心肌保护作用机制。方法Wistar大鼠随机分为对照组、模型组、生脉组及生脉硒高、中、低剂量组,通过腹腔注射异丙肾上腺素制备大鼠急性心肌缺血损伤模型。生脉组以10 mg/(kg.d)量灌胃生脉注射液,对照组与模型组予以等量生理盐水,生脉硒高、中、低剂量组分别按20、10、5 mg/(kg.d)灌胃生脉硒水溶液。观察各组大鼠心肌组织结构的改变,测定血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,免疫组化法观察凋亡蛋白Bcl-2和Bax的表达。结果生脉组和生脉硒组可明显降低心肌梗死面积,同时提高血清SOD活性、降低MDA含量(P<0.05)。免疫组化显示,生脉硒组Bcl-2蛋白阳性表达显著增加(P<0.01),生脉硒中、高剂量组与生脉组明显抑制Bax蛋白表达(P<0.01)。结论生脉硒组对急性心肌缺血损伤模型心肌具有保护作用,其机制可能与抗氧化作用、对抗心肌细胞的凋亡有关。
Objective To observe the effect of Shengmai Selenium on myocardial ischemic injury rats induced by isoproterenol(ISO).Methods Wistar rats were randomly divided into control group,model group,Shengmai group and high-,medium-,low-dose of Shengmai Selenium group.Acute myocardial ischemic injury model was made by intraperitoneal injection of ISO.Shengmai group was given Shengnai Injection 10 mg/(kg.d) by gavage,control group and model group were given equal volume NS,and high-,medium-,low-dose of Shengmai Selenium group was given mixture of Shengmai injection and selenium-enriched malt extraction 20,10,5 mg/(kg.d).Myocardial tissue change in the structure was observed.SOD activity and MDA content were determined and the expression of apoptotic protein Bax/Bcl-2 was observed by immunohistochemistry method.Results Shengmai group and Shengmai Selenium group significantly reduced myocardial infarct size,and elevated serum SOD activity(P 0.05),decreased MDA content(P 0.05).Immunohistochemistry showed that positive expression of Bcl-2 protein in Shengmai Selenium group increased significantly(P 0.01).High-,medium-dose of Shengmai Selenium group and Shengmai group significantly inhibited the expression of Bax protein(P 0.01).Conclusion Shengmai Selenium has myocardial protective effect on acute myocardial ischemic injury rat model.Its mechanism may be related with its antioxidant effect and against myocardial cell apoptosis.
