摘要:目的:观察菖蒲郁金汤对多发性抽动症(tourette syndrome,TS)模型大鼠多巴胺(dopamine,DA)代谢酶[儿茶酚胺氧位甲基转移酶(catechol-O-methyltransferase,COMT)、单胺氧化酶B(monamine oxidase B,MAO-B)]的影响,探讨TS发病与COMT和MAO-B的关系以及菖蒲郁金汤对COMT和MAO-B的影响。方法:将72只雄性SD大鼠随机分为空白组12只,造模组60只,造模组大鼠腹腔注射亚氨基二丙腈(Iminodipropiononitrile,IDPN)制备TS动物模型,空白组大鼠腹腔注射等体积生理盐水,1次/d,连续7天,造模后造模组大鼠表现出不同程度的运动行为及刻板行为,评分均≥2分,表明造模成功。造模成功后将造模组大鼠分为模型组、泰必利组及菖蒲郁金汤高、中、低剂量组,各12只。从造模完第2天开始灌胃,泰必利组给予泰必利(3.194 mg/mL)混悬液,菖蒲郁金汤高、中、低剂量组分别给予相应浓度的菖蒲郁金汤(5.152、2.576、1.288 g生药/mL)药液,空白组与模型组给予等体积纯净水,每日1次,连续给药4周。最后1次给药后24 h采集标本,用WB法、RT-PCR法检测大鼠纹状体COMT、MAO-B蛋白及mRNA表达。结果:各组大鼠纹状体COMT、MAO-B蛋白及mRNA表达差异无统计学意义(P>0.05),但菖蒲郁金汤各剂量组显示出与其剂量相关的COMT表达增强的趋势。结论:1)TS发病可能与COMT和MAO-B无明显关系;2)菖蒲郁金汤的抗抽动作用可能与MAO-B无明显关系,而与COMT表达的增强有一定关系。
Objective:To observe the influence of ChangPu YuJin decoction on DA metabolic enzymes[catechol-O-methyltransferase(COMT),monamine oxidase B(MAO-B)]of the rat models with Tourette's syndrome(TS),and discuss the relationship between the incidence of TS and COMT,MAO-B,and the effects of ChangPu YuJin decoction on COMT and MAO-B.Methods:All 72 male SD rats were randomized into 12 ones in the blank group and 60 ones in the modeling group,the rats of the modeling group accepted peritoneal injection of Imino-dipropiononitrile(IDPN)to prepare TS animal models,the rats in the blank group received peritoneal injection of physiological saline in the equivalent volume,once each day,for seven days consecutively,after modeling,the rats in the model group performed motor behavior and stereotyped behavior to different degrees,the scores all≥two points,which showed that the models were successfully made.After successfully modeling,the rats in the modeling group were assigned into the model group,tiapride group,high,moderate and low dosages groups of ChangPu YuJin decoction,12 rats each group.Since the second day after modeling intragastric administration began,tiapride group was given tiapride suspension(3.194 mg/mL),high,moderate and low dosages groups of ChangPu YuJin decoction were given the decoction in the corresponding concentrations respectively(5.152,2.576 and 1.288 crude drug/mL),the blank group and the model group accepted the equivalent volume of purified water,once each day,medicating for four weeks consecutively.The samples were collected over 24 hours after the last medication,the expressions of COMT,MAO-B protein and mRNA in rat corpus striatum were detected using WB method and RT-PCR method.Results:The difference had no statistical meaning in the expressions of COMT,MAO-B protein and mRNA in rat corpus striatum among different groups(P>0.05),while different doses groups of ChangPu YuJin decoction showed the tedency that the increase of COMT expressions was related to the dose.Conclusion:1)The attack of TS might have no obvious correlation with COMT and MAO-B;2)Anti-spasm effects of ChangPu YuJin decoction might have no obvious connections to MAO-B,while it could enhance the expressions of COMT to a certain degree.
