摘要:骨关节炎(osteoarthritis,OA)属于骨科临床常见的慢性、增龄性疾病,炎症和软骨退变在其发病机制中起着关键作用。近年来,随着对中医药研究的不断深入,运用中医药防治OA在临床上被广泛应用。活血化瘀类中药能够有效抑制炎症反应,延缓软骨退变,有望成为防治骨关节炎的潜在药物资源。当归是常见的活血化瘀中药,其在骨关节炎的治疗中使用频率较高。药理学研究表明,活性成分阿魏酸(ferulic acid,FA)、当归多糖(Angelica sinensis polysaccharide,ASP)、藁本内酯(ligustilide,LIG)能够有效抑制炎症反应,抑制细胞外基质降解,调节软骨细胞的代谢和阻碍软骨细胞的凋亡,对OA有显著的疗效。该文就当归活性成分在骨关节炎防治领域取得的最新进展与突破做一综述,旨在系统梳理当归活性成分治疗骨关节炎的药理学机制及作用价值,为进一步的基础研究与药物研发提供科学依据。
Osteoarthritis(OA)is a common chronic age-related orthopedic disease.And inflammation and cartilage degeneration play a key role in its pathogenesis.In recent years,with the deepening of researches on Chinese medicine,Chinese medicine has been widely applied for managing OA.Blood-activating and blood-stasis-removing herbal medications may effectively arrest nflammation and delay cartilage degeneration.It offers potential drug resources for preventing and treating OA.Angelica sinensis is a common blood-activating and stasis-eliminating herb.Pharmacological studies have demonstrated that the active ingredients of ferulic acid(FA),Angelica sinensis polysaccharide(ASP)and ligustilide(LIG)could effectively treat OA through suppressing inflammatory responses,arresting extracellular matrix degradation,regulating chondrocyte metabolism and impeding cartilage apoptosis of chondrocytes.This review summarized the latest advances and breakthroughs for ASP in the prevention and treatment of OA.It was intended to provide scientific rationales for further basic researches and drug developments.
