摘要:目的:考察蛇葡萄素钠单用及与卡铂合用对人肺腺癌GLC-82细胞增值的抑制作用,初步探讨蛇葡萄素钠抑瘤作用机制。方法:应用MTT法考察蛇葡萄素钠单用及与卡铂合用对人肺腺癌GLC-82细胞的细胞毒作用;使用流式细胞仪检测凋亡相关基因Bax以及Bcl-2表达。结果:MTT实验结果表明,蛇葡萄素钠单独应用及与卡铂联合应用对GLC-82细胞的增殖均具有浓度依赖性的抑制作用;流式细胞仪检测结果表明,蛇葡萄素钠(100、50、25μg/ml)单用组及与卡铂25μg/ml联合用药组作用于GLC-82细胞48 h后,Bax表达上调,且有浓度依赖性;AMP-Na 50μg/ml单独用药组、卡铂25μg/ml单独用药组以及AMP-Na(50、25、12.5μg/ml)与卡铂25μg/ml联合用药组作用于GLC-82细胞48 h后,Bcl-2蛋白的表达下调,但无浓度依赖性。AMP-Na各剂量组(100、50、25、12.5μg/ml)单用组及AMP-Na各剂量组与卡铂25μg/ml联合用药Bax/Bcl-2的比率增高,且有浓度依赖性。结论:蛇葡萄素钠单用对GLC-82细胞的增殖具有一定的抑制作用,与卡铂合用,对GLC-82细胞增殖具有协同抑制效应;蛇葡萄素钠可能是通过下调Bcl-2并上调Bax从而促进肿瘤细胞凋亡。
To investigate the cytotoxic effect and mechanism of ampelopsin sodium on human lung adenocarcionma cell line GLC-82 by alone or combined with carboplatin. Methods: Use the MTF colorimetric method to investigate cytotoxicity effect of ampelopsin sodium alone or combined with carboplatin to human lung adenocarcionma cell llne GLC-82 ; Use flow cytomctry(FCM) analysis the express of Bax and Bcl- 2. Results: Experimental study on cytotoxicity in vitro showed that, ampelopsin sodium( 100, 50, 25 μg/ml) alone or combined with carbo- platin 25 μg/ml all had concentration dependent depressant effect to the generation of GLC-82 cell; FCM results showed that, 48 hours after ampelopsin sodium( 100, 50, 25, 12.5 μg/ml)combined with carboplatin25 μg/ml to GLC-82 cell, the express of Bax concentration de- pendent up-regulation; the express of Bcl-2 in ampelopsin sodium(50 μ/ml) group, carboplatin 25μ/ml group, ampelopsin sodium(50, 25, 12.5 μg/ml ) combined with carboplatin 25 μg/ml group down regulation, but there, s no concentration dependent; the proportion of Bax/Bcl-2 raise up in all the group. Conclusion: Ampelopsin sodium had depressant effect to the generation of GLC-82 cell, There is a syn- ergistic cytotoxic effect on GLC-82 cells treated with ampelopsin sodium combined with carboplatin. The mechanism of ampelopsin sodium is probably that it can down regulation the express of Bcl-2 and up-regulation express of Bax.
