摘要:目的探讨郁金散对抗生素相关性大肠湿热证(A-DHS)模型大鼠Th17/Treg表达的影响。方法将60只SD大鼠随机分为正常组、模型组、柳氮磺吡啶组、郁金散高、中、低剂量组。利用人工气候箱模拟"外湿、外热"环境,以生物因素和抗生素应激诱导建立抗生素相关性大肠湿热证大鼠模型,观测各组大鼠体温、肠道敏感性和粪便性状变化,ELISA法检测血清中Th17(IL-6、IL-17)及Treg(IL-10、TGF-β)相关细胞因子水平,HE染色法观察结肠组织病理学变化,免疫组化法检测结肠组织中RORγt、Foxp3蛋白表达水平。结果 2.2~9 g/kg郁金散可明显缓解A-DHS模型大鼠发热、腹痛及腹泻症状,降低血清IL-17、IL-6、TGF-β水平并提高IL-10水平,增强结肠组织中Foxp3蛋白的表达并抑制RORγt蛋白的表达,改善结肠组织病理学变化。结论Th17/Treg免疫失衡与A-DHS的发病过程密切相关,郁金散对A-DHS有一定的治疗作用,纠正Th17/Treg免疫失衡,缓解免疫反应对肠道组织的损伤是其作用机制之一。
This study was performed to investigate the influence of Yujin power on Th17/Treg immune imbalance in antibiotic-related large intestine dampness-heat syndrome (A-DHS)rat model.The rat model was established by multiple factors.Total of sixty SD rats were randomly divided into normal control group,model group, positive control group (Sulfasalazine),high,middle and low dose of Yujin power groups,with 10 rats in each group. Seven days after antibiotic treatment,anal temperature and abdominal withdrawal reflex were observed,and the parting grade of faeces shape was scored.The defecation frequency and stool water content were also measured. Enzyme-linked immunosorbent assay (ELISA)was used to measure the levels of Th17 (IL-6,IL-17) and Treg (IL- 10,TGF-β)in serum of rats.Immunohistochemical method was used to detect the expression of RORγ/t and Foxp3 protein in colonic tissues.The pathological changes of colonic tissues were also observed by HE staining. Data showed that 2.2-9.0g/kg Yujin power could significantly improve the general condition of A-DHS rats:the anal temperature and the score of abdominal withdrawal reflex were decreased,and the symptoms of diarrhoea were also reduced.Comparison with model group,the levels of IL-17,IL-6 and TGF-β in serum were decreased,but the level of IL-10 was increased in Yujin power treated rats.The results of immunohistochemical staining showed that Yujin power could up-regulate the expression of Foxp3 and down-regulate the expression of RORγt,while HE staining showed that Yujin power could improve the pathological changes of colonic tissues.In conclusion,the immune imbalance of Th 17/Treg may play an important role in the pathogenesis of A-DHS.Yujin powder has certain therapeutic effect on A-DHS,and one of the mechanisms is regulating the balance of Th17/Treg expression and alleviating the damage of the immune response to the intestinal tissue.
