摘要:目的探讨红芪总黄酮对高强度离心运动大鼠耐力及骨骼肌中一氧化氮(NO)和一氧化氮合酶(NOS)的影响。方法将72只Wistar大鼠随机分为安静对照组、离心运动组和药物预处理组,各组再依据运动前后时间点不同分为运动前1h,运动后1,24,48h4个亚组,每组6只。离心运动前3d,药物预处理组大鼠每日定时给予0.2mg/kg红芪总黄酮灌胃,其余各组大鼠给予等量生理盐水灌胃,观察大鼠最长离心运动时间。离心运动后,取各组大鼠腓肠肌,制备匀浆液,检测NO含量及NOS活性。结果药物预处理组运动时间为(86.25±7.97)min,与离心运动组的(73.12±5.23)min比较差异有统计意义(P<0.05)。离心运动后1h,离心运动组大鼠骨骼肌中NO含量急剧下降(P<0.01),运动后24h,NO含量开始回升,至运动后48h,NO含量接近正常水平;红芪总黄酮可减缓离心运动后大鼠骨骼肌中NO含量的下降(P<0.05),对NOS的表达及活性没有影响。结论红芪总黄酮可增强高强度离心运动大鼠耐力,其机制可能与其减缓骨骼肌中NO含量的急剧下降有关。
Objective To explore the effects of total flavonoids of Hedysarum polybotrys on exercise tolerance,nitric oxide(NO)and nitric oxide synthase(NOS)in skeletal muscle of rats after intensity eccentric exercise.Methods 72 Wistar rats were randomly divided into control group,eccentric exercise group,drug pretreatment group.All groups were then randomized into 1h-subgroup before exercise,1h-,24h-and 48h-subgroups after e xercise respectively,6rats in each subgroup.Rats in the drug pretreatment group were administered total flavonoids of Hedysarum polybotrys(0.2mg/kg)orally on 3days before the centrifugal movement,and the rats in the other groups were given the same amount of normal saline,once a day.The longest eccentric exercise time was observed.Gastrocnemius muscle of rats in all groups was obtained at different time points after the centrifugal movement.NO and NOS in the gastrocnemius muscle were measured by spectroscopy respectively.Results The running time in pretreatment group and eccentric exercise group were(86.25±7.97)min and(73.12±5.23)min respectively,there was a significant difference(P<0.05).After eccentric exercise for 1h,the content of NO was significantly declined in rat gastrocnemius muscle(P<0.01),and close to normal level after 48 hours.Total flavonoids of Hedysa-rum polybotrys could slow down the decline of NO(P<0.05),and has no effect on the expression and activity of NOS.Conclusion Total flavonoids of Hedysarum polybotrys can enhance exercise tolerance of centrifugal movement rats.The mechanism may be related to slowdown the decline of NO in skeletal muscle.
