摘要:目的:通过研究红芪多糖对db/db小鼠糖尿病心肌病心肌组织中TGF-β1与Collagen Ⅰ表达的影响,探讨其对db/db小鼠心肌纤维化的作用。方法:将60只6周龄雄性db/db小鼠随机分为5组:模型组、红芪多糖200mg/kg、100mg/kg、50mg/kg剂量组和罗格列酮4mg/kg组;正常组为12只同周龄同背景的非转基因雄性db/m小鼠。连续灌胃干预8周,于给药前及给药后第2、4、6、8周末检测各组小鼠空腹血糖及体重;Masson染色观察小鼠左心室心肌胶原纤维化程度;Western blot、RT-PCR法检测心肌组织TGF-β1与Collagen I的蛋白和m RNA表达。结果:干预治疗8周后,红芪多糖200mg/kg、100mg/kg剂量组和罗格列酮4mg/kg组血糖较模型组明显下降;Masson染色可见亮绿色胶原纤维在模型组心肌细胞间及血管周围较正常组明显增多且呈堆积状分布;模型组TGF-β1与Collagen I蛋白及m RNA表达水平与红芪多糖50mg/kg剂量组无显著差异,其余各治疗组较模型组明显降低,其中红芪多糖200mg/kg剂量组和罗格列酮4mg/kg组改善更明显。结论:红芪多糖对db/db小鼠心肌纤维化具有保护作用,其作用机制可能是通过抑制TGF-β1的异常表达而降低Collagen I降解实现的。
Objective: To discuss the effects of Polysaccharide from Hedysarum polybotrys Hand.-mazz (HPS) on myocardial fibrosis in mice through the study of HPS on expressions of TGF-β1 and Collagen I in myocardial tissue of db/db mice with diabetic cardiomyopathy(DCM). Method: 60 male db/db mice about 6 weeks old were randomly divided into 5 groups: model group, HPS 200mg/kg,100mg/kg,50mg/kg dose groups,rosiglitazone 4mg/kg group, while 12 non transgenic male db/m mice with the same age and background were as normal group. After the continuous administration of 8 weeks, the fasting blood glucose and weight of all the mice were detected before and after the adminis- tration of 2,4,6,8 weeks, the degree of left ventricular myocard-ial collagen fibrosis of mice was determined by Masson staining. Western blot and RT-PCR were used to detect the expression of TGF-β1 ,Collagen I protein and mRNA in cardiac muscle tissue. Results: After 8 weeks of administration treatment, compared to model group, the blood glucose of HPS 200mg/kg, 100mg/kg dose groups as well as rosiglitazone 4mg/ kg group significantly decreased ( P 〈 0.05 ). The result of Masson staining showed that bright green collagen fibers were heaped up between myocardial cells and around blood vessels. The expression of TGF-β1, Collagen I protein and mRNA had no significant change( P 〉 0.05 ) in model group and HPS 50mg/kg dose groups, while they significantly decreased in the other groups ( P 〈 0.05 ) , especially the 200mg/kg HPS and 4mg/kg rosiglitazone significantly improved them ( P 〈 0.05 ). Conclusion: HPS has a protective effect on myocardial fibrosis in db/db mice,and its mechanism may be achieved by inhibiting the abnormal expression of TGF-β1 and reducing the degradation of Collagen I.
