摘要:目的探讨红芪多糖对急性放射性肺炎小鼠的保护作用及其机制。方法小鼠随机分为空白组、模型组、吡非尼酮组(200 mg/kg)、红芪水煎液组(5000 mg/kg)和红芪多糖低、中、高剂量组(15、30、60 mg/kg),每组24只。预防给药7 d后,除空白组外,其余各组小鼠照射16 Gy全胸X线进行造模,继续给药7 d。各组于造模前进行CT影像学检测,造模后第6天进行肺功能及CT影像学检测。最后一次给药30 min后,采用ELISA法检测血清TNF-α、IL-6、MDA水平及SOD、GSH-Px活性,HE染色观察肺组织病理形态变化,Western blot法检测肺组织TNF-α、mTOR、HIF-1α、VEGF蛋白表达,RT-qPCR法检测肺组织mTOR、VEGF、HIF-1αmRNA表达。结果红芪多糖可在一定程度上改善急性放射性肺炎小鼠的精神状态、活动量、皮毛光泽。与模型组比较,红芪多糖高剂量组小鼠每分钟通气量增加(P<0.01),小鼠肺脏CT值和肺系数降低(P<0.05,P<0.01);红芪多糖中、高剂量组小鼠血清TNF-α、IL-6、MDA水平降低(P<0.05,P<0.01),SOD、GSH-Px活性升高(P<0.05,P<0.01),肺组织病理炎性反应减轻,肺泡炎病理评分降低(P<0.05,P<0.01),肺组织mTOR、VEGF、HIF-1αmRNA表达和TNF-α、mTOR、HIF-1α、VEGF蛋白表达降低(P<0.05,P<0.01)。结论红芪多糖可能通过调控HIF-1信号通路来防治小鼠急性放射性肺炎的发生发展。
AIM To explore the protective effect and mechanism of Hedysarum polybotrys polysaccharides on mouse model of acute radiation pneumonia.METHODS The mice were randomly divided into the blank group,the model group,the pirfenidone group(200 mg/kg),H.polybotrys decoction group(5000 mg/kg),and low,medium and high dose H.polybotrys polysaccharides groups(15,30,60 mg/kg),with 24 mice in each group.All of the mice,except those of the blank group,were exposed to 16 Gy full chest X-ray for modeling following 7 days of preventive administration,who then resumed another 7 days corresponding administration.The mice had their CT imaging tests prior to the modeling,and lung function and CT imaging tests on the 6th day after modeling as well.Thirty minutes after the last administration,the mice had their the levels of serum TNF-α,IL-6,MDA and the activities of SOD and GSH-Px detected by ELISA;their pulmonary pathological changes observed by HE staining;their pulmonary protein expressions of TNF-α,mTOR,VEGF and HIF-1αdetected by Western blot;and their pulmonary mRNA expressions of mTOR,VEGF and HIF-1αdetected by RT-qPCR.RESULTS H.polybotrys polysaccharides somehow improved the mental state,activity and fur luster of mouse models of acute radiation pneumonia.Compared with the model group,the high dose H.polybotrys polysaccharides group demonstrated increased ventilation per minute(P<0.01),and decreased lung CT value and lung coefficient(P<0.05,P<0.01).And the medium and high dose H.polybotrys polysaccharides groups displayed decreased levels of serum TNF-α,IL-6,MDA(P<0.05,P<0.01);increased activities of SOD and GSH-Px(P<0.05,P<0.01);alleviated pulmonary pathological inflammatory reaction,decreased pathological score of alveolitis(P<0.05,P<0.01);and decreased pulmonary expressions of mTOR,VEGF and HIF-1αmRNA and TNF-α,mTOR,VEGF and HIF-1αproteins(P<0.05,P<0.01).CONCLUSION H.polybotrys polysaccharides may prevent and treat acute radiation pneumonia in mice via HIF-1 signaling pathway.
