摘要:目的探讨当归补血汤及其加味方对大鼠脑缺血再灌注损伤的神经保护作用及其机制。方法采用线栓法制成大鼠大脑中动脉缺血再灌注模型,免疫组化染色法检测大鼠脑缺血再灌注后海马区NGF阳性细胞的表达,TTC染色法体现大鼠脑组织梗死体积的改变,观察当归补血汤及其加味方对其结果的影响。结果脑缺血再灌注及生理盐水灌胃组大鼠海马区NGF阳性细胞分布稀疏,数目较假手术对照组、脑缺血再灌注后当归补血汤灌胃组及当归补血汤加味方灌胃组明显减少(P<0.05);大鼠脑缺血再灌注后当归补血汤灌胃组及当归补血汤加味方灌胃组相对生理盐水灌胃组均能减少大鼠脑缺血再灌注损伤后的脑组织梗死体积(P<0.05)。结论当归补血汤及其加味方能增加大鼠脑缺血再灌注后海马区NGF阳性细胞的表达,同时减少脑缺血再灌注后大鼠的脑组织梗死体积,对大鼠脑缺血再灌注后的神经组织具有一定的保护作用。
Objective To investigate the Danggui (Angelica sinensis) complex mixture and its additional prescriptions" neuropro- tection function on the rat cerebral ischemia - reperfusion. Methods Using thread - tie method to make middle cerebral artery is- chemia - reperfusion model,immunohistochemical staining for NGF detection of focal cerebral ischemia - reperfusion in rat hip- pocampus expression of cells, the TTC staining method to observe the brain infarction volume change and the Danggui complex mixture and its additional prescription influence their results. Results Hippoeampus of cerebral ischemia reperfusion and intraper- itoneal fed of saline group NGF - immunoreactive cells have sparse distribution, compared with sham - operated control group, the number of cerebral ischemia reperfusion after intraperitoneal fed of the Danggui complex mixture and its additional prescription group significantly reduced ( P 〈 0.05 ) ; and the rat cerebral ischemia - reperfusion after by intraperitoneal fed of the Danggui complex and its additional prescription group relative cerebral ischemia reperfusion of intraperitoneal fed group all can reduce rats after ischemia - reperfusion injury of cerebral infarction(P 〈 0.05 ). Conclusion The Danggui comples mixture and its additional prescription can enhance the expression of NGF - positive ceils in hippocamus after, cerebral ischemia reperfusion, at the same time reduce the rat cerebral infaction volume of brain tissue,which has a protective effect on ischemia reperfusion.
