摘要:目的:分析比较甘肃礼县掌叶大黄水根与掌叶大黄药用部位活性成分的差异。方法:采用正交试验法,以乙醇浓度、乙醇用量、提取时间和提取次数为考察因素,芦荟大黄素、大黄素、大黄酸3个成分提取量为考察指标,借助高效液相色谱(HPLC)法测定,通过综合加权评分法进行数据分析。结果:大黄及水根最佳提取工艺参数为每次加4倍量75%乙醇,回流提取3次,每次60min;对大黄及水根3种成分的综合评分进行双样本方差分析,结果无显著性差异。结论:在优选的提取工艺下,大黄水根与大黄药用部位活性成分的含量并无显著性差异。该结论可为进一步有效开发水根,提高其利用价值提供依据。
Objective:To analyze and compare difference between antibacterial components of branch and main roots of Rhubarb.Methods:Orthogonal experimental method was adopted to optimize extraction conditions.Ethanol concentration,ethanol dosage,ethanol extraction time and extraction times were taken as influencing factors,and aloe emodin,emodin and rhein contents were taken as evaluation indexes.HPLC was used to determine biomarker contents and data were dealt with weighted scoring method.Results:The optimal extraction conditions were:75%ethanol was used as extraction solvent,and extraction times were selected as three and each time was 60 min.Double sample variance analysis was conducted for the difference in the three components contents of branch and main roots of Rhubarb.There was not significant variance between branch and main roots of Rhubarb based on three biomarkers analyzed in this study.Conclusion:Under the optimal extraction conditions,the content of three biomarkers of branch and main roots of Rhubarb is not significant.The conclusion can provide theoretical basis for effectively developing branch root of Rhubarb,and increasing its utilization value.
