摘要:目的:研究补阳还五汤预处理对心肌缺血再灌注损伤(MIRI)大鼠心肌酶及抗氧化功能的影响。方法:将40只Wistar大鼠随机分为假手术组(Sham组)、缺血再灌注组(MIRI组)、缺血预适应组、补阳还五汤组各10只。造模前,4组大鼠分别连续灌胃7 d,补阳还五汤组用补阳还五汤灌胃,其余各组用等量生理盐水灌胃,采用结扎冠状动脉左前降支(LAD)方法复制MIRI大鼠模型。通过大鼠心电图评判是否造模成功,采用生化法检测血清乳酸脱氢酶(lactate dehydroge-nase,LDH)和肌酸激酶(creatine kinase,CK)的活性,采用硫代巴比妥酸法测定血清丙二醛(malondialdehyde,MDA)含量,黄嘌呤氧化酶法测定血清超氧化物歧化酶(superoxide dismutase,SOD)活性及硝酸还原酶法测定一氧化氮(Nitric oxide,NO)含量。结果:与MIRI模型组比较,补阳还五汤组血清LDH和CK活性明显降低,MDA含量明显降低,SOD活性及NO含量明显升高。结论:补阳还五汤可提高MIRI大鼠血清中SOD活性,增加NO含量,同时降低LDH、CK活性和MDA含量,减轻MIRI,其机制与减轻细胞膜脂质过氧化反应,防护再灌注期心肌细胞的氧化应激损伤有关。
Objective:To study the effects of Buyang Huanwu Decoction pretreatment on myocardial enzymes and antioxidant functions in myocardial ischemia reperfusion injury(MIRI)rats.Methods:40 Wistar rats were randomly divided into Sham group(Sham group),ischemia reperfusion group(MIRI group),ischemia preconditioning group and Buyang Huanwu Decoction group.Before the modeling,the rats in the four groups were given continuous gavage for 7d,the rats in the Buyang Huanwu Decoction group were given gavage with Buyang Huanwu decoction,and the other groups were given normal saline,and the MIRI rat model was replicated by ligating the left anterior descending coronary artery(LAD)method.ECG was used to evaluate the success of rats modeling,biochemical method was used to detect the serum lactate dehydrogenase(LDH)and creatine kinase(CK)activity.The determination of malondialdehyde(MDA)in serum was measured by thiobarbituric acid method.Serum superoxide dismutase(SOD)activity was measured by xanthine oxidase method.NO content was measured by Nitrate reductase method.Results:compared with the MIRI model group,serum LDH and CK activity in Buyang Huanwu Decoction group decreased significantly.MDA concentration was significantly decreased,SOD activity and NO content were significantly increased.Conclusion:Buyang Huanwu Decoction can increase SOD activity,increase NO content,decrease LDH,CK activity and MDA concentration in serum of MIRI rats,and reduce MIRI.Its mechanism is related to reducing lipid peroxidation reaction of cell membrane and protecting myocardial cells from oxidative stress injury during reperfusion.
