摘要:目的建立湿生扁蕾乙醇提取物中总[口山]酮的含量测定方法,并对该法的测量不确定度进行评定。方法采用紫外分光光度法测定湿生扁蕾乙醇提取物中总[口山]酮的含量,并进行方法学验证。建立数学模型,识别和分析不确定度来源,量化各不确定度分量,计算测量结果的合成不确定度和95%置信区间下的扩展不确定度。结果在95%置信区间下,紫外分光光度法测得湿生扁蕾乙醇提取物中总[口山]酮的含量为(12.87±0.52)%。供试品溶液中总[口山]酮质量浓度、实验重复性和加样回收率的不确定度是影响总的不确定度的3个主要不确定度分量。结论建立了紫外分光光度法测定湿生扁蕾乙醇提取物中总[口山]酮含量的方法。通过不确定度评定,得到了测量结果在95%置信区间下的扩展不确定度,了解了该法测定结果的可靠性,也为优化该分析方法,提高分析结果质量提供了依据。
Objective To establish a method for the content determination of total xanthones in ethanol extracts from Gentianopsis paludosa and to evaluate the uncertainty in measurement using the method.MethodsUltraviolet spectrophotometry was used to determine the content of total xanthones in ethanol extracts from Gentianopsis paludosa,and the method was verified. The mathematical model was established,the sources of uncertainty were identified and analyzed,the uncertainty components were quantified,and the combined uncertainty of measurement results and the expanded uncertainty in 95 % confidence interval were calculated as well.ResultsWithin 95 %confidence interval the total xanthones content in the ethanol extracts from Gentianopsis paludosa measured by ultraviolet spectrophotometry was(12.87 ± 0.52)%. The uncertainty of concentration of total xanthones in the testsolution, the uncertainty of test repeatability, and the uncertainty of sample recovery rate were the three mainuncertainty components that affect the total uncertainty.ConclusionA method for the content determination of totalxanthones in ethanol extracts of Gentianopsis paludosa by ultraviolet spectrophotometry was established. By evaluatingthe uncertainty, the expanded uncertainty of measurement results at 95% confidence interval was obtained, thereliability of the measurement results of the method was realized, and a basis was provided for optimizing theanalytical method and improving the quality of the analytical results.
