摘要:目的正交实验优选唐古特大黄茎的提取工艺。方法选用L9(34)正交表,以提取次数、提取时间、液固比为考察因素,以岀膏率、粗多糖、苹果酸的含量为考察指标,优化唐古特大黄茎的提取工艺参数。结果唐古特大黄茎的最佳提取工艺为12倍量蒸馏水,煎煮法提取3次,每次1 h,该工艺条件下能获得收率、质量稳定的唐古特大黄茎提取物,平均岀膏率为38.65%,多糖含量为6.64%,苹果酸含量为17.28%。结论优化的工艺简单、稳定、可行,重复性好,可用于唐古特大黄茎的提取。
Objective To optimize the extraction process of the stems of Rheum tanguticum Maxim.ex Balf.Methods The L9(34)orthogonal table was used,with the extracting solvent volume,number of extracting operation and extraction times as related factors,and the yield of total extract and the contents of crude polysaccharide and malic acid in the extract were used as evaluation index for optimizing the extraction processes of the stems of R.tanguticum.Results The conditions for the optimized extraction process of the stems of R.tanguticum were as follows:the ratio of distilled water(ml)to the stems(g)is 12∶1(ml/g),extracting three times by decocting for 1 hour for each time.Using this process,the stem extract of R.tanguticum was obtained with good yield and stable quality.The average extraction yield was 38.65%17.28%.Conclusion The optimized process is simple,stable,feasible,and repeatable,which might be used for the extraction of the stems of R.tanguticum.
