摘要:目的:筛选秦艽愈伤组织缓慢生长保存的适宜方法,进一步研究保存后愈伤组织形态指标与生理指标之间的相关性。方法:向培养基中添加不同浓度生长抑制剂、高渗化合物及不同基本培养基保存秦艽愈伤组织,在室温下保存6个月后统计愈伤组织的形态学指标。测定不同浓度甘露醇、矮壮素和蔗糖处理中秦艽愈伤组织的生理指标。结果:1.0mg·L-1ABA和9%蔗糖处理中愈伤表面产生大量胚状体,生长良好。5%甘露醇和PEG处理中愈伤黄绿色,生长良好。PP333中愈伤生长一般,CCC中愈伤老化。9%蔗糖处理中可溶性糖、淀粉和可溶性蛋白质含量明显高于其它浓度和甘露醇、矮壮素处理。结论:1.0mg·L-1ABA,9%蔗糖保存秦艽愈伤组织效果最好;5%甘露醇和PEG保存效果良好。可溶性糖、淀粉和可溶性蛋白质3个生理指标可以在一定程度上反映秦艽愈伤组织在延缓保存时的存活情况。
Objective : An appropriate conservation method for callus of Gentiana macrophylla Pall. slowly growing, and the relationship of physiological index and morphological index of the conserved callus were studied. Methods: To conserve callus, growth inhibitors and high permeability compounds were added to the medium, or different concentration of basic medium were used. After the callus was conserved for six month at room temperature, the morphological index was recorded and physiological index of the conserved callus of Gentiana macrophylla with different concentration of mannitol, CCC and sucrose were measured. Results: A large number of embryoid were generated in the surface of callus with the treatment of 1.0 mg/L ABA and 9% sucrose and grew well. The callus showed yellow-green and grew well under the treatment with 5% mannitol and PEG. Callus in PP333 grew commonly and callus in CCC was aging. Under the treatment with 9% sucrose, soluble sugar, starch and soluble protein content were significantly higher than treated with the other concentration of sucrose, mannitol or CCC. Conclusion: The best condition for callus conservation of Gentiana macrophyUa was 1.0 mg/L ABA and 9% sucrose, followed by 5% mannitol and PEG. Soluble sugar, starch and soluble protein can partly reflect the survival rate of the callus conservation slowly growing.
