摘要:目的研究五脉绿绒蒿总黄酮和总生物碱抗肝纤维化的作用。方法皮下注射CCl4、喂饲高脂低蛋白质复合饲料并饮用20%乙醇来复制肝纤维化大鼠模型,通过HE染色观测肝组织病理变化,测定大鼠肝脏指数、肝组织羟脯氨酸以及血清肝纤维化指标。结果五脉绿绒蒿醇提物、总黄酮及其与总生物碱的混合物能增加大鼠的体质量,降低肝脏指数、肝组织羟脯氨酸与胶原蛋白的水平以及肝纤维化指标(透明质酸、Ⅲ型前胶原、Ⅳ型胶原和板层素),改善肝纤维化模型大鼠肝组织病理变化,其中五脉绿绒蒿总黄酮的抗肝纤维化作用显示出一定的量效关系。结论五脉绿绒蒿醇提物及其中的总黄酮具有一定的抗肝纤维化作用。
AIM To observe and compare the protective effect of total flavonoids and alkaloids from Meconopsis quintuplinervia on rat with hepatic fibrosis.METHODS The hepatic fibrosis model rats were established by subcutaneously injecing CCl4,fed high-fat low-protein diet and drunk 20% alcohol.Ethanol extract,total flavonoids and total alkaloids from Meconopsis quintuplinervia were used to detect the levels of hydroxyproline(Hyp),collagen fiber in hepatic tissue and liver pathology.RESULTS 90% ethanolic extract,total flavonoids and mixture of total flavonoids and total alkaloids from Meconopsis quintuplinervia could increase the body weight of experimental rats,decrease the serum levels of hepatic fibrosis(hyaluronic acid,precollagen Ⅲ,collagen Ⅳ and laminin) and of hydroxyproline,collagen protein in hepatic tissue,and improve liver pathology.And the effect of total flavonoids exhibited the dose-effect relationship.CONCLUSION 90% ethanolic extract and total flavonoids from Meconopsis quintuplinervia have some effect on the hepatic fibrosis in rats.
