摘要:目的观察当归挥发油对脑缺血大鼠血液流变学的影响及防止血栓形成的作用。方法将60只大鼠随机分成6组,分别为假手术组,模型组,当归挥发油大、中、小剂量组及阳性对照组,灌胃给药7 d,每天2次,于末次给药1 h后采用大脑中动脉阻断术(MCAO)制作大鼠脑缺血模型,测定血液流变学各项指标;通过小鼠尾静脉注射胶原蛋白与肾上腺素混合诱导剂诱导血栓形成,观察5 min内小鼠的死亡数和15 min内小鼠偏瘫的恢复数。结果当归挥发油能显著降低大鼠的全血黏度、全血还原黏度以及红细胞压积、红细胞刚性指数,对血浆黏度、红细胞聚集指数、红细胞变形指数无明显影响;同时当归挥发油可减少注射血栓诱导剂后小鼠的死亡数,增加小鼠偏瘫恢复数。结论当归挥发油可显著改善全血黏度等血液流变学多项指标,并能有效防止血栓的形成。
Objective To observe the effects of Angelica naphtha on blood rheology of rats with cerebral in- farction and the effect of preventing thrombosis. Methods 60 rats were randomly divided into 6 groups : sham ope- ration group, model group, high-dose, middle-dose, low-dose of Angelica naphtha group, Ginkgo leaf positive control group. All rats had been administrated with corresponding drug for 7 days, 2 times per day, the rat models with cere- bral infarction were made by adopting MCAO after 1 hour the last administration, and detected each index of blood rheology in blood. Through injecting mixed inducing agent of collagen and epinephrine in caudal veinto of mice to induce thrombosis, and observed death number of mice in 5 minutes and recovery number of hemiplegia of mice in 15 minutes. Results Angelica naphtha could significantly reduce blood viscosity, whole blood reduced viscosity and hematocrit, red blood cell rigidity index, no significant effects on plasma viscosity, red blood cell aggreagtion index and red blood cell distortion index. At the same time, Angelica naphtha could significantly reduce death number of mice in 5 minutes after injecting mixed inducing agent, and induce recovery number of hemiplegia of mice in 15 minutes. Conclusion Angelica naphtha can significantly improve many index of blood rheology, and prevent effectively thrombosis.
