摘要:目的:应用来曲唑建立多囊卵巢综合征(PCOS)大鼠模型,探讨当归水提物对来曲唑(Letrozole)致大鼠多囊卵巢的保护作用及其作用机制。方法:60只SD大鼠随机分为正常对照组、模型对照组、二甲双胍0.3 g/kg组、当归水提物2 g/kg、4 g/kg、8 g/kg组,每组10只。正常对照组每天上午灌胃1%羧甲基纤维素钠溶液,其余组灌胃含来曲唑(1 mg/kg)的1%羧甲基纤维素钠溶液,灌胃体积为8 mL/kg,连续28 d,建立PCOS大鼠模型。造模同时,各组灌胃给予相应药物或蒸馏水,每日1次,连续28 d。禁食不禁水,16 h后眼球取血,并收集卵巢。采用苏木素-伊红(HE)染色观察卵巢组织的病理变化程度,酶联免疫吸附法(ELISA)测定血清中睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)以及肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的含量,生化法检测卵巢组织中超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px)活性以及丙二醛(MDA)的含量,蛋白免疫印迹法(Western blot)检测Janus蛋白酪氨酸激酶2(JAK2)、磷酸化Janus蛋白酪氨酸激酶2(p-JAK2)、信号转导和转录活化因子3(STAT3)、磷酸化信号转导和转录活化因子3(p-STAT3)的蛋白表达情况。结果:与正常对照组相比,模型对照组大鼠的体重明显增加,血清中T、LH、FSH、TNF-α、IL-1β、IL-6含量显著升高(P<0.01),卵巢组织中SOD、GSH-Px活性显著降低(P<0.01),MDA含量显著升高(P<0.01);与模型对照组相比,当归水提物4 g/kg、8 g/kg组HE染色可见颗粒细胞层数增多,囊状扩张不同程度好转;血清中T、FSH、LH、TNF-α、IL-1β、IL-6含量显著降低(P<0.01),卵巢组织中SOD、GSH-Px活性显著升高(P<0.01),MDA含量显著降低(P<0.01);p-JAK2、p-STAT3蛋白表达显著下调(P<0.01)。结论:当归水提物对来曲唑致多囊卵巢大鼠具有保护作用,主要通过降低激素水平和炎症反应,其机制可能与IL-6介导的JAK2/STAT3信号通路相关。
Objective: To investigate the protective effect and its mechanism of aqueous extract from Angelica sinensis on rat model of polycystic ovary syndrome(PCOS) that was induced by letrozole. Methods: 60 SD rats were randomly divided into the control group, the model group, 0.3 g/kg metformin group, 2 g/kg, 4 g/kg and 8 g/kg aqueous extract from Angelica sinensis groups, with 10 rats in each group. 1% sodium carboxymethyl cellulose solution was intragastrically administered in the control group every morning, while 1% sodium carboxymethyl cellulose solution containing letrazol(1 mg/kg) was intragastrically administered in other groups, 8 mL/kg each day, for 28 consecutive days, to establish the PCOS rat model. After 16 hours of fasting with water only, blood was taken from the eyeball and ovaries were collected. The pathological changes of ovarian tissues were observed by hematoxylin-eosin(HE) staining. The contents of testosterone(T), luteinizing hormone(LH), follicle-stimulating hormone(FSH) and tumor necrosis factor alpha(TNF-α), interleukin-1 beta(IL-1β) and interleukin-6(IL-6) in serum were measured by ELISA. The activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-PX) and the content of malonaldehyde(MDA) in ovarian tissues were detected by biochemical method. Western blot was performed to detect the protein expressions of Janus tyrosine kinase 2(JAK2), phosphorylated Janus tyrosine kinase 2(p-JAK2), signal transduction and transcriptional activation factor 3(STAT3), phosphorylated signal transduction and transcriptional activation factor 3(p-STAT3). Results: Compared with the contol group, the levels of T, LH, FSH, MDA, TNF-α, IL-1β and IL-6 in the model group were significantly increased(P<0.01), while the activities of SOD and GSH-PX were significantly decreased. Compared with the model group, the contents of T, LH, FSH, MDA, TNF-α, IL-1β and IL-6 in 4 g/kg and 8 g/kg aqueous extract from Angelica sinensis groups were significantly decreased, and the activities of SOD and GSH-PX were significantly increased. The protein expressions of p-JAK2 and p-STAT3 in the ovarian tissue were significantly decreased. The result of HE staining showed that the number of granule cell layers was increased and the cystic expansion was improved to different degrees. Conclusion: The aqueous extract from Angelica sinensis has a protective effect on rat with polycystic ovary syndrome induced by letrozole, mainly through reducing hormone levels and inflammatory responses, and its mechanism may be related to the JAK2/STAT3 signaling pathway mediated by IL-6.
