摘要:目的探讨当归对阴虚哮喘小鼠的平喘作用及对肺组织中水通道蛋白5(AQP5)表达的影响。方法通过注射卵清白蛋白(OVA)致敏、吸入OVA激发的方法复制哮喘小鼠模型,实验后期给予甲状腺素复制阴虚哮喘模型,观测哮喘发作及其程度、肺功能、肺液清除功能,血清白介素-13(IL-13)、α-肿瘤坏死因子(TNF-α)的水平和肺组织中AQP5及其基因的表达。结果当归能抑制哮喘发作并缓解其症状,改善肺功能,促进肺组织AQP5及其基因的表达,降低血清IL-13与TNF-α的水平;当归与地塞米松配伍后,对改善肺功能、调节AQP5及其基因表达等有一定的协同作用。结论当归具有一定的平喘作用,调节AQP5及基因表达进而促进肺组织水代谢平衡是其作用机制之一。
OBJECTIVE To investigate the effects of Angelicae sinensis Radix(ASR) on anti-asthma and the expression of aquaporin-5(AQP5) in lung tissue of asthmatic mice with Yin deficiency syndrome.METHODS Ovalbumin(OVA) was injected to sensitize,OVA was inhaled to stimulate the mice,and Thyroxin was used during late stimulation.The asthmatic mice model with Yin deficiency syndrome was established and evaluated through asthmatic attack and its degree,pulmonary function,pulmonary fluid clearance,IL-13 and TNF-α in the serum,and expression of AQP5 and its gene in the lung tissue.RESULTS ASR could reduce the asthmatic attack and its degree,improve pulmonary function,depress IL-13 and TNF-α in serum,and promote the expression of AQP5 and AQP5 mRNA in the lung tissue.In addition,these is a certain synergy between ASR and Dexamethasone on improving pulmonary function and the expression of AQP5 and its gene.CONCLUSION ASR has anti-asthmatic effect,and one of mechanisms is to regulate the expression of AQP5 and AQP5 mRNA to promote water metabolism balance.
