摘要:目的观察痛泻要方对溃疡性结肠炎(UC)模型大鼠结肠黏膜ERK1、ERK2基因表达的影响,探讨其作用机制。方法将实验动物分为6组,采用TNBS/乙醇灌肠法造模,肉眼观察结肠大体形态损伤并进行评分,以大鼠结肠黏膜ERK1、ERK2为观察指标,采用RT-PCR法检测ERK1、ERK2基因表达水平。结果肉眼观察模型组大鼠结肠组织黏膜层可见炎症和溃疡形成,与空白组比较,P<0.05,证实模型成功。模型组大鼠结肠组织ERK1、ERK2基因的相对表达量均高于空白组(P<0.05);治疗后,痛泻要方高剂量组、中剂量组ERK1、ERK2基因的相对表达量均较模型组升高(P<0.05,P<0.01)。结论痛泻要方对TNBS/乙醇法UC大鼠模型结肠黏膜ERKl、ERK2基因的相对表达量有上调作用,提示痛泻要方治疗作用可能与ERK信号转导途径被激活有关。
OBJECTIVE To observe the influence of Tongxie Yao Fang on the ERK1 and ERK2 gene expressions of colonic mucosa in rats with ulcerative colitis (UC),and to identify its underlying mechanism of action.METHODS Experimental animals were divided into six groups and prepared by means of TNBS/ ethanol enema to observe their morphological injury and damage with the naked eyes,and to detect ERK1 and ERK2 gene expressions by means of RT-PCR with ERK1 and ERK2 being the observation targets.RESULTS Inflammation and ulceration on the colonic mucous membrane of rats were observed with naked eyes,which suggested a successful modeling compared with the control group (P<0.05).The relative expression amount of ERK1 and ERK2 of colonic tissues of rats in the model group was higher than that of the blank group (P<0.05) ;after treatment,compared with the model group,the relative expression amount of ERK1 and ERK1 in the prescription highand moderate-dose groups were increased (P<0.05 and P<0.01).CONCLUSION Tongxie Yao Fang can promote the relative expression amount of ERK1 and ERK2 of colonic tissues of rats with UC,which suggests the therapeutic effects of the prescription are probably related to the activation of ERK signal transduction pathway.
