摘要:目的:建立赤芍不同提取部位高效液相色谱(HPLC)指纹图谱,探讨其抑制ras突变肿瘤活性的谱效关系,为明确其抗肿瘤作用及物质基础提供参考。方法:分别用石油醚、醋酸乙酯、正丁醇对赤芍水煎液进行萃取,并采用HPLC法对提取物进行分析,建立色谱图;采用秀丽隐杆线虫检测各有效部位的急性毒性及抗肿瘤活性;采用偏最小二乘(PLS)回归分析研究其谱效关系。结果:赤芍醋酸乙酯部位的急性毒性高于正丁醇部位和石油醚部位;赤芍3个部位均不同程度地提高了线虫的野生型比例,且正丁醇部位效果最好;谱效关系研究表明,色谱峰7、5(芍药苷)、1所对应化合物对抑制ras突变的肿瘤活性有重要作用。结论:赤芍抑制ras突变的抗肿瘤活性是多种化学成分共同作用的结果,为赤芍作为抗肿瘤药物的开发研究提供了一定思路。
OBJECTIVE To establish high performance liquid chromatography(HPLC) fingerprints of different extract parts from Paeoniae Rubra Radix,to explore the spectrum-effect relationship between extracts and anti-tumor activity of inhibiting ras mutation,and to provide references for clarifying its anti-tumor activity and material basis.METHODS The extracts of Paeoniae Rubra Radix were extracted with petroleum ether,ethyl acetate and n-butanol,and were analyzed by HPLC to establish its chromatograms.The acute toxicity and anti-tumor activity of each effective part were detected by C.elegans.The spectrum-effect relationship was studied by partial least squares(PLS) regression analysis.RESULTS The acute toxicity of the ethyl acetate fraction was higher than that of n-butanol and petroleum ether.The three parts of Paeoniae Rubra Radix all increased the proportion of wild type of nematodes to varying degrees,but the n-butanol part had the best effect.The study of the spectrum-effect relationship showed that the compounds corresponding to peaks 7,5(Paeoniflorin) and 1 played an important role in inhibiting the tumor activity of ras mutations.CONCLUSION The anti-tumor activity of inhibiting ras mutation was the result of a combination of various chemical components,which will provide a idea for the development of Paeoniae Rubra Radix as an anti-tumor drug.
