摘要:目的 探讨桃红四物汤加味对银屑病小鼠Th17/Treg平衡及卵巢雌二醇分泌的干预作用。方法 将72只SPF级雌性BALB/c小鼠随机分为空白组、模型组、阳性对照组、桃红四物汤加味低、中、高剂量组,通过在小鼠背部涂抹5%咪喹莫特乳膏诱导建立小鼠银屑病样皮肤损伤模型,并在建模同时进行药物干预。采用HE、TUNEL、IHC、ELISA、流式、Western blot和Real-time PCR等实验技术检测相关指标。结果 与模型组比较,各给药组小鼠PASI评分、Baker评分、皮损组织ki-67阳性表达量和卵巢组织颗粒细胞凋亡百分率降低,IL-17浓度降低,IL-2、IL-10、E2、P450scc、3β-HSD、17β-HSD浓度升高,P450scc、3β-HSD、17β-HSD mRNA表达水平升高,外周血中Th17+细胞阳性率和RORγt蛋白表达水平降低,Treg+细胞阳性率和Foxp3、ERα、ERβ蛋白表达水平升高,Th17/Treg细胞比例降低,差异显著(P<0.05)。结论 桃红四物汤加味可明显改善卵巢颗粒细胞凋亡,促进E2分泌及其受体ERα、ERβ表达,减轻炎性细胞浸润和表皮增厚程度,通过抑制Th17细胞分化,促进Treg细胞分化,调节Th17/Treg平衡,起到治疗银屑病的作用。
Objective To explore the intervention effect of Taohong Siwu Decoction Jiawei(桃红四物汤加味,TSDJ)on Th17/Treg balance and ovarian estradiol(E2)secretion in psoriasis mice.Methods Seventy-two SPF female BALB/c mice were randomly divided into the blank group,model group,positive control group,TSDJ low/medium/high-dose groups.The psoriasis-like skin lesions model of mice was induced by applying 5%imiquimod cream on the back of mice,and drug intervention was given at the same time of modeling.Relevant indicators were detected by HE staining,TUNEL assay,IHC staining,ELISA,flow cytometry(FCM),Western blot(WB),and Real-time PCR(qPCR).Results Compared with the model group,PASI and Baker score,Ki-67 positive expression level in skin lesion tissue,and granulosa cell apoptosis rate in ovarian tissue were decreased in all treatment groups,while the concentration of IL-17 was decreased,and concentrations of IL-2,IL-10,E2,P450scc,3β-HSD,and 17β-HSD were increased.The mRNA expression levels of P450scc,3β-HSD,and 17β-HSD were elevated.In peripheral blood,the positive rates of Th17+cells and the protein expression level of RORγt were decreased,while the positive rate of Treg+cells and the protein expression levels of Foxp3,ERα,and ERβwere increased,resulting in a reduced Th17/Treg cell ratio,with significant differences(P<0.05).Conclusion TSDJ can significantly alleviate ovarian granulosa cell apoptosis,promote the secretion of E2 and the expression of its receptors ERαand ERβ,reduce the degree of inflammatory cell infiltration and epidermal thickening,suppress Th17 cell differentiation,promote Treg cell differentiation,regulate Th17/Treg balance,and thereby exerting a therapeutic effect against psoriasis.
