摘要:目的观察黄芩苷对H22肝癌小鼠瘤组织磷脂酰肌醇-3-羟激酶(PI3K)、AKT和哺乳动物雷帕霉素靶蛋白(mTOR)表达的影响。方法60只昆明小鼠随机分为肿瘤模型组(模型组)、环磷酰胺组(环组)、黄芩苷低、中、高剂量组(黄低组、黄中组、黄高组)及黄芩苷中剂量联合环磷酰胺组(联合组),每组10只,各组给予相应药物灌胃或腹腔注射,连续14 d。处死小鼠后测量瘤体质量计算抑瘤率,实时荧光定量PCR技术检测瘤组织PI3K、AKT和mTOR基因表达,免疫组织化学方法检测小鼠瘤组织PI3K、AKT和mTOR蛋白表达。结果与模型组比较,各给药组瘤体质量均下降(P<0.05),黄低组、黄中组、黄高组和联合组抑瘤率≥50%,各给药组PI3K、AKT和mTOR基因和蛋白表达均降低(P<0.05),尤其黄高组和联合组蛋白表达降低更明显(P<0.01)。结论黄芩苷可抑制肝癌荷瘤小鼠瘤组织的增长,其作用机制可能与下调瘤组织PI3K、AKT和m TOR基因及蛋白表达有关。
Objective To observe the effects of baicalin on the expression of PI3 K,AKT and mTOR in tumor tissue of H22 Hepatoma-bearing mice.Methods Sixty Kun-ming mice were randomly divided into tumor model group(model group),cyclophosphamide group(CTX group),baicalin low,medium and high-dosage groups(low group,middle group and high group)and baicalin medium-dose combined with cyclophosphamide group(combined group),10 rats in each group.The mice were administered with corresponding drugs for 14 d.The mice were executed,the tumor tissue were taken and weighed,the tumor inhibition rate was calculated.The gene expressions of PI3 K,AKT and mTOR in tumor tissues was detected by real-time quantitative PCR technology.The protein expressions of PI3 K,AKT and mTOR in tumor tissues were detected by immunohistochemistry.Results Compared with the model group,the tumor mass of each administration group decreased.The tumor inhibition rate of low group,middle group,high group and combination group was≥50%.The levels of PI3 K,AKT and mTOR gene and protein expression in each administration group were decreased,especially the protein expression in high group and combined group.Conclusion Baicalin can inhibit the growth of tumor tissues in H22 hepatoma-bearing mice by adjusting PI3 K,AKT and mTOR gene and protein expression at both levels.
