摘要:目的研究红芪多糖对大鼠肝纤维化的药效作用。方法 Wistar大鼠用二甲基亚硝胺(DMN)建立肝纤维化模型。将大鼠随机分为正常组、模型组、红芪多糖高、中、低剂量组、黄芪多糖组、红芪水煎液组。采用酶联免疫法检测血清谷丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST),肝组织Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C)、羟脯氨酸(Hyp)、透明质酸(HA)、层黏连蛋白(LN)含量,HE和Masson染色观察肝组织形态变化,测定胶原纤维面积百分比。结果与模型组比较,红芪多糖各组ALT、AST、HA、Hyp、LN、PCⅢ、Ⅳ-C含量均明显降低(P<0.05或P<0.01),镜下观察肝纤维化明显减轻,胶原纤维面积明显减少(P<0.05或P<0.01)。结论红芪多糖能明显改善肝组织纤维化,具有抗肝纤维化作用。
Objective To study the pharmacodynamic effect of Hedysarum polybotrys saccharide (HPS) on rats' hepatic fibrosis. Methods The hepatic fibrosis models (HFM) were composed of Wistar rats which were intraperitoneally injected with dimethylnitrosamine (DMN) periodically. Wistar rats were divided into 7 groups: normal control, model, high, middle and low dose HPS treatment, astragalus polysaccharides treatment, hedysari elixation groups, in accordance with the principles of randomization. At the end of experiment, type m procollagen (PCⅢ), type IVcollagen (IV-C), hydroxyproline oxyprolinc (Hyp) , hyaluronic acid (HA) and liminin (LN) content of he- patic tissue were detected by ELISA. Hepatic tissues were taken for the observation in morphological transmutation in HE and Masson stai- ning method to determine the collagen fiber area percentage. Results For the HPS treated rats, HA, Hyp, LN, PC m, IV-c levels in liver tissues were obviously lower than those of the HFM group (P 〈0. 05 or P 〈 0. 01 ) , it could be seen that the degree of hepatic fibrosis was decreased obviously examined with microscope, the area of Collagen fibers was reduced obviously ( P 〈0. 05 or P 〈 0.01 ) , the difference was significant. Conclusions HPS can improve the degree of hepatic fibrosis and play an anti-hepatic fibrosis role.
