摘要:目的探讨当归多糖对哮喘幼龄小鼠的平喘作用及对抗氧化系统的影响。方法按照体重将小鼠随机分为5组:正常组、模型组以及高、中、低3个剂量实验组(当归多糖:240,120,60 mg·kg^(-1)),每组10只。利用卵清白蛋白复制哮喘模型。用肺功能仪测定呼气峰(PEF),酶联免疫吸附法测定支气管肺泡灌洗液(BALF)中超氧化物歧化酶(SOD)和过氧化氢酶的活性。结果正常组、模型组和高、中、低3个剂量实验组的PEF分别为(115.48±28.16),(59.02±12.81),(85.37±17.38),(78.69±16.24)和(72.03±15.92)mL·s^(-1);这5组的SOD活性分别为(74.58±7.96),(47.61±5.76),(81.83±9.09),(71.67±7.62)和(63.14±6.57)U·mg^(-1);这5组的过氧化氢酶活性分别为(15.87±1.70),(7.79±0.86),(16.59±1.77),(14.67±1.58)和(13.02±1.35)U·mg^(-1)。上述指标模型组与正常组比较,差异均有统计学意义(均P<0.01);3个剂量实验组与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论当归多糖对幼龄小鼠哮喘具有改善作用,而提高抗氧化系统的活性是其作用机制之一。
Objective To investigate the effects of Angelica Sinensis polysaccharide(ASP) on anti-asthma and antioxidant system in young asthmatic mice. Methods The young mice were divided into 5 groups according to weight: normal group, model group and high, medium and low dose experimental groups(240,120,60 mg·kg^(-1) ASP). The peak expiratory flow(PEF)was determined by Spirometer. The activities of superoxide dismutase(SOD)and catalase in bronchoalveolar lavage fluid were observed by enzyme linked immunosorbent assay. Results The PEF of normal group, model group and high, medium and low dose experimental groups were(115.48±28.16),(59.02±12.81),(85.37±17.38),(78.69±16.24)and(72.03±15.92)mL·s^(-1);the SOD in the 5 groups were(74.58±7.96),(47.61±5.76),(81.83±9.09),(71.67±7.62)and(63.14±6.57)U·mg^(-1);the catalase in the 5 groups were(15.87±1.70),(7.79±0.86),(16.59±1.77),(14.67±1.58)and(13.02±1.35)U·mg^(-1). Comparison between model group and normal group, the difference of the factors were significant(all P<0.01);comparison between three doses experimental groups and model group, the difference of the factors were significant(P<0.01). Conclusion Angelica sinensis polysaccharide has the improvement achived effect on asthma in young mice,and enhancing the activity of antioxidant system is one of its mechanisms.
