摘要:目的:比较大黄不同品种、不同产地加工品蛋吉、苏吉和水根的蒽醌含量。方法:采用RP-HPLC法测定大黄不同种类不同产地加工品中蒽醌的含量。色谱柱:Hypersil ODS-2 C18(4.6 mm×250 mm,5μm);Kromasil ODS保护柱(4.6 mm×10mm,5μm);流动相:甲醇-0.1%磷酸(85∶15);流速:0.8 mL.min-1;检测波长:435 nm;柱温:25℃。结果:大黄不同品种、不同规格的产地加工品,其蒽醌含量存在很大差异。无论是蛋吉、苏吉和水根,以四川药用大黄蒽醌含量为最高,甘肃礼县掌叶大黄次之,再次为青海和四川若尔盖唐古特大黄,甘肃渭源和陇西掌叶大黄为差;对同一品种、不同规格的产地加工品的蒽醌含量,蛋吉≥苏吉>水根。结论:大黄不同品种、不同规格的产地加工品,其蒽醌含量存在很大差异。不同品种大黄其蒽醌含量:药用大黄>掌叶大黄>唐古特大黄;不同规格加工品:蛋吉≥苏吉>水根。主根明显优于支根。
Objective:To compare the anthraquinone contents in different processed products and species of Rhu- barb including Danji, Suji and Shuigen. Methods: The anthraquinone content was determined by RP - HPLC. The separation was performed on a Hypersil ODS -2 C18 column(4.6 mm ×250mm,5μm) and a Kromasil ODS guard column(4. 6 mm×10mm,5μm) eluted with the mobile phase of methanol -0. 1% phosphoric acid(85: 15) at the flow rate of 0. 8 mL · min ^- l, and the wavelength was set at 435 nm and 25 ℃ for column. Results : There were differences in anthraquinone contents among different species and processed products of Rhubarb in different produ- cing areas. The content of Rheum officinale Baill. from Sichuan was the highest, Rheum palmatum L. from Gansu Lixian ranked the second, and Rheum tanguticum Maxim. ex Ball. from Sichuan and Qinghai was the third, and Rheum palmatum L. from Gansu Weiyuan and Longxi were the lowest. The rank of the content of different processed products of Rhubarb was listed as the follow :Danji was the highest, Suji was the second, and Shuigen was the low- est. Conclusion:The anthraquinone content varies in different species and processed products of Rhubarb were as followed respectively :Rheum officinale 〉 Rheum palmatum 〉 Rheum tanguticum ; Danji ≥ Suji 〉 Shuigen. The con- tent in main roots was significantly higher than that in the lateral roots.
