摘要:目的:探讨芍药苷对大鼠慢性溃疡性结肠炎的干预机制。方法:采用葡聚糖硫酸钠(DSS)诱导慢性溃疡性结肠炎模型,30只Wistar大鼠正常饮水饮食7d,第8天给予添加2%DSS的蒸馏水自由饮用7 d,无添加的蒸馏水连续饮用14 d,上述DSS/蒸馏水循环饮用3个循环。将模型大鼠随机区组法分为模型组、芍药苷组、柳氮磺吡啶组,每组为10只大鼠,另取10只大鼠设为空白对照组,给予生理盐水代替DSS。ELISA法检测外周血TNF-α、IL-6的含量;免疫组织化学、免疫印迹法检测自噬相关蛋白Beclin1(BECN1基因)、抗凋亡蛋白Bcl-2(B淋巴细胞瘤-2基因)的表达。结果:与空白对照组相比较,模型组大鼠外周血TNF-α、IL-6含量显著升高(P<0.05),Beclin1表达显著降低(P<0.05),Bcl-2表达增加(P<0.05)。与模型组相比较,芍药苷、柳氮磺吡啶处理后外周血TNF-α、IL-6含量显著降低(P<0.05),Beclin1表达显著增加(P<0.05),Bcl-2表达显著降低(P<0.05)。结论:慢性溃疡性结肠炎存在自噬表达水平减弱或缺失,芍药苷能够下调抗凋亡基因Bcl-2表达,上调自噬相关蛋白Beclin1表达,减轻DSS诱导的慢性溃疡性结肠炎。
Objective:To explore the intervention mechanism of Paeoniflorin in alleviating chronic ulcerative colitis in rats.Methods:Chronic ulcerative colitis models were induced by using dextran sulfate sodium(DSS),30 Wistar rats were fed with normal diet for 7 days.On the 8 th day,the 2%DSS was added to the distilled water for free drinking 7 days,and then no added distilled water for drinking 14 days,and the above DSS/distilled water circulated for 3 cycles.After the models were made successfully,30 rats were divided into model group,Paeoniflorin group and Sulfasalazine positive control group by randomized block design,with 10 rats in each group.In addition,another 10 rats were taken as normal control group and given the physiological saline instead of the DSS.The levels of TNF-α and IL-6 in peripheral blood were measured by using ELISA,the expressions of autophagy related protein Beclin1 and anti-apoptotic gene Bcl-2 were detected by immunohistochemistry and Western Blot.Results:Compared with the normal control group,the contents of TNF-α and IL-6 in peripheral blood were significantly increased(P<0.05),while the expression of Beclin1 was significantly decreased(P<0.05),the expression of Bcl-2 was significantly increased(P<0.05).Compared with the model group,the contents of TNF-α and IL-6 were reduced,meanwhile the expression of Beclin1 was enhanced(P<0.05)and the expression of Bcl-2 was lowered(P<0.05).Conclusion:The expression of autophagy was weakened or absent in chronic ulcerative colitis,Paeoniflorin can reduce the intestinal inflammation induced by DSS,its mechanism may be related to down-regulation the expression of anti-apoptotic gene Bcl-2 and up-regulation the expression of autophagy associated protein Beclin1.
