摘要:目的研究射干苷对银屑病小鼠中Th17/Treg平衡的影响。方法小鼠涂抹5%咪喹莫特乳膏建造银屑病模型,随机分为正常组、模型组、低剂量射干苷组(2.5 mg/kg)、高剂量射干苷组(10 mg/kg)和甲氨蝶呤组(2.5 mg/kg)。HE染色观察皮肤组织病理变化,免疫组化法检测Bax、Bcl-2、Ki67蛋白表达,ELISA法检测血清TNF-α、IL-6、IL-10水平,流式细胞术检测外周血Th17/Treg细胞比例,RT-qPCR检测组织miRNA-204-5 p、JAK 2、STAT 3 mRNA表达,Western blot检测组织Bax、Bcl-2、JAK2、p-JAK2、STAT3、p-STAT3蛋白表达。结果射干苷可改善银屑病小鼠皮损区皮肤病理变化,抑制生发层细胞和角质形成细胞的异常增殖,促进其凋亡;同时可减轻银屑病小鼠的炎症反应,降低血清中TNF-α、IL-6、IL-17A水平,降低外周血Th17/Treg比例,提高银屑病小鼠皮损区皮肤中miRNA-204-5 p表达,抑制JAK2和STAT3的mRNA和蛋白表达,以及二者磷酸化蛋白表达。结论射干苷可提高银屑病小鼠皮损区皮肤组织中miRNA-204-5 p表达,抑制JAK2/STAT3信号通路的异常活化,减轻炎症反应,进而通过平衡角质形成细胞的增殖与凋亡,改善银屑病小鼠皮肤银屑病症状。
AIM To investigate the effects of tectoridin on the balance of Th17/Treg in mice with psoriasis.METHODS Unlike the mice of the normal control group for no intervention,other mice developed into psoriasis models with 5%imiquimod cream treatment were subsequently randomly divided into model group,groups for low-dose or high-dose tectoridin administration(2.5,10 mg/kg),and methotrexate group(2.5 mg/kg).The mice had their skin histopathological changes observed by HE staining;their protein expressions of Bax,Bcl-2 and Ki67 detected by immunohistochemistry;their serum levels of TNF-α,IL-6 and IL-10 detected by ELISA;their ratio of Th17/Treg cells in peripheral blood determined by flow cytometry;their mRNA expressions of miRNA-204-5 p,JAK 2 and STAT 3 in tissues investigated by RT-qPCR;and their protein expressions of Bax,Bcl-2,JAK2,p-JAK2,STAT3 and p-STAT3 in tissues detected by Western blot.RESULTS Tectoridin improved the pathological changes of psoriatic skin lesions,inhibited the abnormal proliferation of germinal layer cells and keratinocytes,and promoted their apoptosis.Additionally,it alleviated the inflammatory reaction;decreased the serum levels of TNF-α,IL-6 and IL-17A,and the ratio of Th17/Treg in inperipheral blood;increased the expression of miRNA-204-5 p in skin lesions;and inhibited the mRNA,protein expressions and the phosphorylated protein expressions of JAK2 and STAT3.CONCLUSION Tectoridin reduces the inflammatory reaction,and improves the skin symptoms of mice with psoriasis by balancing the proliferation and apoptosis of keratinocytes via increase of miRNA-204-5 p expression of skin lesions,inhibition of the abnormal activation of JAK2/STAT3 signaling pathway.
