摘要：目的观察当归腹痛宁滴丸(当归油)对肠易激综合征模型大鼠的治疗作用,探讨作用机制。方法采用8种刺激诱导大鼠建立肠易激综合征模型,随机分为正常组、模型组、阳性组(匹维溴铵)、当归腹痛宁滴丸高、中、低剂量组,每组10只,连续给药31 d,定期测定各组大鼠的体质量、摄食量、排便情况、自主运动量,末次给药后测定胃排空率和肠推进率,放射免疫法测定血清以及结肠匀浆5-HT、血浆和结肠匀浆P物质(SP)、血管活性肠肽(VIP)水平。结果给予当归腹痛宁滴丸后,大鼠摄食量和排便量增加,固体粒状粪便增多,随后体质量和摄食明显增加,运动量接近正常,依次为当归腹痛宁滴丸高、中剂量组。大鼠胃排空率增加和肠推进率减小亦如此。当归腹痛宁滴丸使大鼠血清5-HT水平下降,血浆VIP水平升高,和结肠匀浆5-HT水平下降,以高剂量组为明显。结论当归腹痛宁滴丸对肠易激综合征具有一定的治疗作用,其作用机制与抑制脑肠轴5-HT和VIP过度分泌和向正常水平恢复有关。
AIM To study the therapeutic effect of Danggui Futongning Dropping Pills（ angelica oil） in rat with irritable bowel syndrome. METHODS Mice＇models for irritable bowel syndrome were induced by eight types of stimulations. Sixty SD rat were divided into the normal control group,the model group,positive control group（ Pinaverium Bromide Tablets）,high-dose,middle-dose,low-dose Danggui Futongning Dropping Pills group. The treatment lasted for 31 days. Body weights,appetite,defecation,independent exercise of all rats were recorded. The rate of gastric emptying and small intestinal propulsion rate were measured. The 5-HT in serum,substance P（ SP） and vasoactive intestinal peptide（ VIP） in plasma or 5-HT,SP,VIP in colon homogenates were evaluated by radioimmunoassay. RESULTS With the treatment of Danggui Futongning Dropping Pills,the food intake and stool quantity increased. Consequently,body weights and appetite increased as well. The independent movement returned to normal level. High-dose Danggui Futongning Dropping Pills group performed the best and followed by middle-dose group. The same results observed in those two groups for emptying rates and intestinal propulsion rates. The 5-HT levels in serum and in colonic homogenate significantly reduced but VIP levels in plasma increased markedly. The SP and VIP contents significantly decreased in colonic homogenate. CONCLUSION The therapeutic effect of Danggui Futongning Dropping Pills on irritable bowel syndrome is obvious. The mechanism could be related to inhibition against the excessive secretion of 5-HT and VIP in brain-gut-axis.
