摘要:目的:对镉染毒(Cd)大鼠给予小补肾汤,探讨小补肾汤对镉细胞免疫功能和氧化应激损伤的影响。方法:选用SPF级Wistar大鼠60只,随机分为正常组、模型组、小补肾汤干预组(高、中、低剂量)、乙酰半胱氨酸组。末次给药24 h后处死大鼠,计算胸脾指数。采用MTT法检测T淋巴细胞的转化能力,比色分析法测定大鼠血清SOD活性和MDA含量,ELISA法测定大鼠血清IL-2和TGF-β_1的含量,免疫组化法检测大鼠Bcl-2和Bax的表达。结果:与正常组比较,模型组大鼠的体质量明显减轻,胸脾指数及T淋巴细胞的刺激指数明显降低,SOD活性明显降低,IL-2含量明显降低,Bcl-2的蛋白表达明显降低,MDA含量和TGF-β_1的含量均明显升高,Bax的蛋白表达明显升高(P〈0.05或P〈0.01)。与模型组比较,小补肾汤中、高剂量组体质量及T淋巴细胞的刺激指数明显增高,Bcl-2的蛋白表达明显升高,高剂量组胸腺指数明显增高,各干预组脾脏指数、SOD活性、IL-2含量明显增高;而小补肾汤中、高剂量组Bax的蛋白表达明显降低,各干预组MDA含量和TGF-β_1的含量明显降低(P〈0.05或P〈0.01)。结论:小补肾汤对镉所致大鼠的免疫损伤和细胞凋亡具有保护作用。
Objective : To study the effects of Dunhuang Prescriptions Small Kidney - Tonifying Decoction on the damage of cell immune function and oxidative stress in rats induced by exposure to cadmium. Methods : Sixty SPF Wistar rats were randomized divided into six groups : normal control group, cadmium - exposed group, four administrated groups. After killing, indexes of thymus and spleen were calculated by weighing method. The transformation ability of T lymphocyte was measured by MTT. The levels of SOD and MDA were detected by colorimetric assay. The contents of IL - 2 and TGF - 1β were analyzed by ELISA. The protein expressions of Bcl - 2 and Bax were tested by immunohistochemical staining. Results : Compared with the normal control group, the weight of cadmium - exposed group was decreased significantly and the SOD activity was decreased significantly, while the MDA level of cadmium - exposed group was increased significantly ( P 〈0.01 ). The thymus and spleen indexes of the cadmium - exposed group were lower than those in the normal control group( P 〈 0.01 ). The IL- 2 content of cadmium -exposed group was lower than those in the normal control group, but the TGF - 1β content of cadmium - exposed group was higher than those in the normal control group (P 〈 0.01 ). The Bcl - 2 protein expression of cadmium - exposed group was lower than those in the normal control group, but the Bax protein expression of cadmium - exposed group was higher than those in the normal control group ( P 〈 0.01 ). Compared with the cadmium - exposed group, the weights and the T lymphocyte stimulation indexs of middle, high dose Small Kidney -Tonifying Decoction groups were increased significantly (P 〈 0.05 or P 〈 0.01 ). The spleen indexes of Small Kidney- Tonifying Decoction treated groups were higher than those in the cadmium - exposed group(P 〈0.05 or P 〈0.01 ). Sim- ultaneously, the thymus index of high dose Small Kidney - Tonifying Decoction group was higher than those in the cadmi- um - exposed group ( P 〈 0.05 ). The MDA levels of Small Kidney - Tonifying Decoction treated groups were lowered sig- nificantly, while the SOD activities were increased significantly (P 〈 0.05 or P 〈 0.01 ). The IL - 2 contents of Small Kidney - Tonifying Decoction administrated groups were increased significantly, and the TGF-1β contents in Small Kid- ney- Tonifying Decoction administrated groups were lower significantly(P 〈 0.05 or P 〈 0.01 ). The Bcl -2 protein ex- pression of middle, high dose Small Kidney - Tonifying Decoction groups were increased significantly, and the Bax protein expression middle, high dose Small Kidney - Tonifying Decoction groups were lower significantly(P 〈 0.05 or P 〈 0.01 ). Conclusion: Small Kidney -Tonifying Decoction has protective effect on immunity function damage and cell apoptosis in- duced by cadmium in rats.
