摘要:目的观察加味黑布膏对大鼠耳廓复合痤疮模型血清IL-8和TNF-α含量的影响,探讨外用加味黑布膏治疗痤疮的作用机制。方法将60只Wistar大鼠随机分为6组,每组10只,取任意1组做空白组,另外5组造模成功后随机分为模型组、阳性对照组、加味黑布膏低、中、高剂量组。空白组、模型组不涂抹任何药物,阳性对照组涂抹0. 025%维A酸乳膏,加味黑布膏低、中、高剂量组涂抹不同剂量的加味黑布膏,1次/d,连续给药4周后,采用ELISA法检测各组大鼠血清IL-8和TNF-α的含量。结果各治疗组大鼠血清IL-8和TNF-α的含量均低于模型组(P <0. 05);低、中剂量组与阳性对照组比较差异无统计学意义(P> 0. 05),高剂量组与阳性对照组比较差异有统计学意义(P <0. 05)。结论加味黑布膏治疗痤疮的作用机制可能与抑制和减少IL-8和TNF-α的产生有关。
Objective To observe the effect of modified Heibu ointment on serum IL-8 and TNF-α levels in rat model with compound acne on the auricle,and to explore the mechanism of external application of modified Heibu ointment in the treatment of acne.Methods Sixty Wistar rats were randomly divided into 6 groups,with 10 rats in each group.One group was selected randomly as a blank group,and the other 5 groups were randomly divided into a model group,a positive control group,and a low,medium and high dose modified Heibu ointment groups after successful modeling.The blank group and the model group were not applied with any drug,the positive control group was applied with 0.025% vitamin A acid cream,and the low,medium and high dose modified Heibu ointment groups were applied with different doses of modified Heibu ointment once per day.After continuous administration of 4 weeks,IL-8 and TNF-α levels in the serum of each group was detected by ELISA.Results IL-8 and TNF-α levels in serum of the rats in each treatment group was lower than that in the model group(P< 0.05).There was no statistical difference between the low and medium dose groups(P> 0.05),and the difference between the high dose group and the positive control group was statistically significant(P< 0.05).Conclusion Mechanism of modified Heibu ointment in the treatment of acne may be related to the inhibition and reduction of IL-8 and TNF-α levels.
