摘要:目的:探讨丹参酮ⅡA联合伊马替尼对K562细胞凋亡作用的影响及相关作用机制。方法:将K562细胞分为6组:常规培养K562细胞组、1μM伊马替尼单药组、20μM丹参酮ⅡA单药组,以及5μM、10μM、20μM丹参酮ⅡA分别与1μM伊马替尼的联合用药组。在显微镜下观察细胞的形态学变化,检测细胞12 h和24 h的生长情况,利用流式细胞仪检测细胞的凋亡情况,再利用JC-1染料在荧光显微镜下观察细胞线粒体膜电位变化情况。结果:丹参酮ⅡA通过抑制线粒体膜电位,促进细胞凋亡(P<0.05);并随丹参酮ⅡA的浓度增加联合用药组的抑制作用也增强。结论:丹参酮ⅡA可显著增强伊马替尼引起的K562细胞凋亡,其机制与线粒体途径有关。
Objective : To investigate the effect of Tanshinone ⅡA with Imatinib on the apoptosis of K562 ceils and its re- lated mechanisms. Methods: K562 cells were cultured and divided into six groups: normal K562 cells group, 20 μM tanshinone ⅡA group, 1 μM Imatinib group, and combined group(5μM, 10 μM, and 20 μM respectively plus 1μM Imatinib). After treatment, the morphological changes of cells were observed under microscope; the cell apoptosis was detected by flow cytometry; and the mitochondrial activity was also observed by using JC - 1 reagent and fluorescence microscope. Results: Tanshinone ⅡA promotes the cell apoptosis by inhibiting the mitochondrial transmembrane poten- tial ( P 〈 0.05 ) ; and the inhibitory effects of tanshinone ⅡA were enhanced with the increase of its concentration. Con- clusion : Tanshinone ⅡA can significantly enhance the apoptosis of K562 treated by Imatinib, which is related with mito- chondrial pathway.
