摘要:目的通过研究当归挥发油对自发性高血压大鼠(SHR)肝脏组织中TRIB1、TLR3 mRNA水平表达的影响,探讨当归挥发油对SHR脂代谢的改善作用及其机制。方法将8周龄雄性SHR随机分为模型组,当归挥发油组低剂量组(含5%的挥发油),当归挥发油组高剂量组(含10%的挥发油)和阿托伐他汀钙组,并用同周龄的Wistar大鼠作为正常对照组。各组大鼠分别灌胃4 w后,测定给药前后血压的变化。取大鼠肝脏组织,HE染色观察各组大鼠肝脏组织的结构变化,QT-PCR检测TRIB1、TLR3 mRNA水平的表达。结果当归挥发油能降低SHR的血压。TRIB1,TLR3在各用药组表达上调,且呈差异性表达,在挥发油组不同剂量组之间亦呈差异性上调表达。结论当归挥发油对SHR脂代谢有调节作用,通过上调TRIB1、TLR3 mRNA水平抑制动脉粥样硬化而降压,从而影响脂代谢。
Objective To observe the Chinese angelica naphtha on liver tissue TRIB1,TLR3 m NRA expression in spontaneously hypertensive rats( SHRs) and explore the effect of angelica naphtha on lipid metabolism and its mechanism.Methods the 8-week-old male SHR were randomly divided into model,low( 5% angelica naphtha),high dose of angelica naphtha( 10% angelica naphtha) and atorvastatin groups,and the Wistar rats with the same age were selected as normal control group. Changes of blood pressure were measured before and after 4 weeks of intragastric administration administration. Liver tissue was taken and HE staining was used to observe the structural changes.Results Angelica naphtha could reduce the blood pressure of SHRs. TRIB1,TLR3 expressions were up-regulated in treatment groups,and there were significant differences between different dose groups. Conclusions Angelica naphtha plays a regulatory role in SHR lipid metabolism by raising the TRIB1,TLR3 mRNA expressions and thus inhibits atherosclerosis and affects lipid metabolism.
