摘要：目的探讨原发性高血压病患者颈动脉硬化与基质金属蛋白酶-3(MMP-3)多态性的关系。方法 104例高血压病患者(高血压病组),健康体检者102例为对照组,MMP-3基因启动子-1171位点产生三种基因型:6A/6A、5A/6A、5A/5A,分析两组基因的分布差异;其中高血压组68例与对照组30例行颈动脉超声检查,观察其内-中膜厚度(IMT)。结果高血压病组MMP-36A/6A基因型和6A等位基因频率均高于对照组(基因型65.4%/53.9%,等位基因82.2%/76.0%),但分布差异无统计学意义(基因型P=0.094,等位基因P=0.120);高血压病组6A/6A基因型者颈动脉分叉前处IMT大于5A/5A+5A/6A基因型者,差异有统计学意义(P<0.05)。结论原发性高血压病患者颈动脉IMT与多种危险因素密切相关,高血压病组6A/6A基因型者颈动脉IMT较厚。
Objective To investigate the association between polymorphism of matrixmetalloproteinase 3 （MMP-3） and cartotid aterty of essential hypertension. Methods This study enrolled 104 patients with hypertension and 102 normal volunteers. After PCR procedure, the genotypes of MMP-3 were scored according to the patterns of DNA bands. The ultrasound examination was performed in 68 patients with hypertension and 30 normal volunteers to investigate the changes of intima- media thickness of carotid arteries.Results The frequency of MMP-3 6A/6A type and allele 6A were higher in the hypertension group than those in the control group（65.4%/53.9%, 82%/62%）,but the difference was not statistical （gtype P=0.094,allele P=0.120）. The IMT of patients with MMP-3 6A/6A gtype was significantly higher compared with that of patients in the control group （P〈0.05）. Conclusion The CCA-IMT is closely associated with many risk actors carotid IMT is higher among patients with 6A/6A gtype.
