摘要:目的筛选能够准确鉴定甘肃省不同肉苁蓉种质的序列,探究甘肃省肉苁蓉种质资源遗传关系。方法利用筛选出的4对引物(ITS2F-ITS3R、CD1F-CD1R、matK3F-matK1R和rbcL1F-rbcL724R)对甘肃省肉苁蓉、沙苁蓉、盐生肉苁蓉和兰州肉苁蓉4种共27份肉苁蓉样品基因组序列进行扩增测序,分析样品的序列特征、核苷酸多态性、单倍型多样性,构建邻接法(NJ)聚类树,从nrDNA和cpDNA两方面进行遗传变异研究。结果cpDNA合并序列GC含量比nrDNA合并序列低,为34.6%;matK序列的变异位点明显高于ITS、CD及rbcL序列,共检测到481个多态性变异位点;两组合并序列单倍型多样性值均大于0.5,多样性程度较高;NJ聚类树显示,nrDNA和cpDNA的合并序列联合使用更适用于肉苁蓉不同种质的鉴别;肉苁蓉遗传变异主要来自组内变异,即同一物种不同种质间分化明显,nrDNA和cpDNA组内变异百分比分别为79.32%和76.93%。结论甘肃省肉苁蓉种质资源遗传多样性较为丰富,nrDNA和cpDNA的合并序列能准确鉴定甘肃省4种肉苁蓉。
Objective To screen the sequences that can accurately identify different Cistanches Herba germplasm in Gansu Province;To explore the genetic relationship of Cistanches Herba germplasm resources.Methods Four pairs of primers(ITS2F-ITS3R,CD1F-CD1R,matK3F-matK1R,and rbcL1F-rbcL724R)were selected to amplify and sequenced 27 samples of 4 Cistanches Herba species(Cistanche deserticola Y.C.Ma,C.sinensis G.Beck,C.salsa(C.A.Mey.)G.Beck,and C.lanzhouensis Z.Y.Zhang)from Gansu Province.Sequence characteristics,nucleotide polymorphism,and haplotype diversity of samples were analyzed,and the neighbor-joining(NJ)clustering tree was constructed.Genetic variation was studied from nrDNA and cpDNA.Results GC content of cpDNA combined sequence was 34.6%lower than that of nrDNA combined sequence.The variable sites of matK were significantly higher than those of ITS,CD,and rbcL,and 481 polymorphism variable sites were detected.The haplotype diversity values of the two combinations were greater than 0.5,indicating a high degree of diversity.NJ clustering tree showed that the combined sequences of nrDNA and cpDNA were more suitable for the identification of different germplasm of Cistanches Herba.Genetic variation of Cistanches Herba was mainly derived from intra-group variation,that was,different germplasm of the same species was differentiated significantly,and the percentage of intra-group variation of nrDNA and cpDNA was 79.32%and 76.93%,respectively.Conclusion Genetic diversity of Cistanches Herba germplasm resources is rich,and the combined sequences of nrDNA and cpDNA can accurately identify 4 Cistanches Herba species in Gansu Province.
