摘要:目的研究黄芪生脉饮(HSY)对放射性心脏损伤(RIHD)炎症反应和纤维化的对抗作用。方法以25Gy X线单次照射大鼠心前区构建RIHD动物模型,HSY低、中、高剂量药物干预组的大鼠照射后每天分别灌胃1.6、3.1、6.2 mL·kg^-1的HSY。通过炎性病理损伤评分,Masson染色心肌胶原定量(CVF),免疫组化和Western Blot检测纤维化因子的表达判断HSY的干预效果。结果模型大鼠照射后1 d、1周、2周、4周、6周病理损伤评分和CVF显著增加(均P<0.01),提示25 Gy X线能引起心脏炎性和纤维化损伤;这种损伤伴随着纤维化因子的高表达,转化生长因子-β1(TGF-β1)、一型胶原(Col1)、三型胶原(Col3)表达呈渐进性升高的趋势,第4周高表达达峰值(均P<0.01)。高、中剂量的HSY可降低照射后大鼠心脏病理损伤评分和CVF(均P<0.01),下调X线引起的纤维化分子的高表达(均P<0.01),从而减弱了放射诱导的心脏炎性-纤维化损伤。结论 HSY可减轻X线照射引起的早期炎性病理反应,下调纤维化分子和胶原纤维的高表达,有效改善照射引起的迟发的纤维化病理进程。
Objective Radiation-induced heart damage(RIHD) is considered to be a comprehensive process of pathological development with progressive inflammatory response and fibrosis.The present study was undertaken to characterize the effect of Huangqi Shengmai yin(HSY) on radiation-induced cardiac inflammatory-fibrosis injury on rat models.Methods Precardiac region of rats were irradiated with 25 Gy X-rays to establish the RIHD model.Low dose(1.6 mL·kg^-1·d^-1,LDG),medium dose(3.1 mL·kg^-1·d^-1,MDG) and high dose(6.2 mL·kg^-1·d^-1,HDG) of HSY were given daily to the irradiated rats.HE staining,Masson staining and collagen volume fraction(CVF)calculating,immunohistochemistry and Western Blot were performed at 1 d,1 w,2 w,4 w,6 w after irradiation.Results The myocardial pathology score of inflammatory injury and collagen volume fraction(CVF) in myocardial tissue increased at 1 d,1 w,2 w,4 w and 6 w after irradiation,indicating that 25 Gy X-rays promoted inflammatory-fibrosis damage effect in rats’ heart.The expression of fibrotic molecules TGF-β1,Col1,Col3 increased gradually after irradiation,and the high expression peaks appeared at 4-w post-irradiation.HSY decreased the score of inflammatory injury and CVF,and decreased the expression of TGF-β1,Coll,Col3 in irradiated rats’ heart,thus alleviated the radiation-induced cardiac inflammatory-fibrosis injury.Conclusion HSY could attenuate the early inflammatory pathological reaction caused by X-ray irradiation,also inhibit the high expression of fibrosis molecules TGF-β1,Col1,Col3,and reduce the collagen fibers deposition in the myocardial tissue.HSY could effectively improve the pathological process of delayed fibrosis caused by irradiation.
