摘要:目的探讨中药复方归芪白术方对大鼠放射性脾损伤的防护作用机制。方法用随机数字表法将大鼠分为3组:正常组、模型组和实验组,每组13只。用6 Gy X线全身照射制作大鼠放射性脾损伤模型。造模前14 d,实验组给予归芪白术方8.4 g·kg-1灌胃,正常组和模型组给予等量的生理盐水灌胃,1次/天。在照射后3 d动物取材,苏木精-伊红染色法观察脾组织的形态变化,免疫组化检测脾组织中核因子κB p56(NF-κB p65)的表达,蛋白质印迹法检测脾组织肿瘤坏死因子受体-1(TNFR1)、核因子κB抑制物激酶β(IKKβ)和环氧合酶-2(COX-2)蛋白表达水平(平均光密度值)。结果模型组大鼠脾组织可见明显的炎性细胞浸润;实验组大鼠脾组织中炎性细胞浸润减少。正常组、模型组和实验组的病理学评分分别为(2.53±0.16),(0.62±0.08)和(1.65±0.11)分;这3组的NF-κB p65蛋白表达水平分别为0.17±0.01,0.26±0.03和0.19±0.01;这3组的TNFR1蛋白表达水平分别为0.15±0.01,0.39±0.01和0.21±0.01;这3组的IKKβ蛋白表达水平分别为0.69±0.01,0.82±0.01和0.68±0.01;这3组的COX-2蛋白表达水平分别为0.55±0.01,0.68±0.01和0.44±0.01。上述指标:模型组与正常组相比,差异均有统计学意义(均P<0.0001);实验组与模型组相比,差异均有统计学意义(均P<0.0001)。结论归芪白术方通过抑制炎症反应发挥对放射性脾损伤的防护作用。
Objective To explore the protective effect and mechanism of Guiqibaizhu Recipe,a traditional Chinese medicine compound,on radiation-induced spleen injury in rats.Methods Sprague-Dawley rats were randomly divided into three groups according to random number table:normal group,model group and experimental group,13 rats in each group.6 Gy X-ray whole irradiation was used to make a rat model of radiation-induced spleen injury.Fourteen days before modeling,the experimental group was given Guiqibaizhu Recipe 8.4 g·kg-1 by gavage,and the normal group and model group were given the same amount of normal saline by gavage,once a day.Three days after irradiation,the animals were collected,and the morphological changes of spleen tissue were observed by hematoxylin-eosin staining method.Immunohistochemical detection of nuclear factor kappa B p65(NF-κB p65)expression in spleen tissue.Western blotting was used to detect the expression levels(average optical density values)of tumor necrosis factor receptor-1(TNFR1),inhibitor of nuclear factor kappa-B kinasebeta(IKKβ)and cyclooxygenase-2(COX-2)proteins in spleen tissue.Results Obvious inflammatory cell infiltration can be seen in the spleen tissue of rats in the model group;inflammatory cell infiltration in the spleen tissue of the experimental group was reduced.The pathological scores of the normal group,model group and experimental group were(2.53±0.16),(0.62±0.08)and(1.65±0.11)point,respectively;the expression levels of NF-κB p65 protein in the three groups were 0.17±0.01,0.26±0.03 and 0.19±0.01,respectively;the expression of TNFR1 protein in the three groups were 0.15±0.01,0.39±0.01 and 0.21±0.01;the expression of IKKβprotein were0.69±0.01,0.82±0.01 and 0.68±0.01,respectively;the expression of COX-2 protein were 0.55±0.01,0.68±0.01 and 0.44±0.01,respectively.Comparison between model group and normal group,the difference of the factors were significant(all P<0.0001);comparison between experimental group and model group,the difference of the factors were significant(all P<0.0001).Conclusion Guiqibaizhu Recipe can protect radiation-induced spleen injury by inhibiting inflammation.
