摘要:目的观察当归贝母苦参丸对顺铂化疗H22荷瘤小鼠肿瘤及肝脏、肾脏和胸腺组织病理形态的影响,探讨其对顺铂化疗的增效减毒作用。方法建立小鼠H22肝癌皮下移植瘤模型,随机分为模型组、顺铂组、当归贝母苦参丸组、当归贝母苦参丸联合顺铂组,另设空白组作对照。造模后第8日开始给药,连续14 d,动态监测荷瘤小鼠肿瘤体积;第15日处死小鼠,镜下观察各组小鼠肿瘤、肝脏、肾脏和胸腺组织的病理形态变化。结果给药第10日,与模型组比较,各给药组肿瘤体积明显减小(P<0.01),当归贝母苦参丸联合顺铂组最为明显;各给药组肿瘤细胞密度、异型性和病理性核分裂像均明显降低(P<0.01),其中当归贝母苦参丸联合顺铂组效果优于顺铂组,还伴有明显的淋巴细胞浸润和纤维组织增生(P<0.01)。顺铂组肝脏、肾脏和胸腺病理变化较空白组、模型组加重(P<0.01),而当归贝母苦参丸联合顺铂组病变较顺铂组减轻(P<0.05)。结论当归贝母苦参丸可以增强顺铂对H22荷瘤小鼠肿瘤的抑制作用,减轻顺铂化疗H22荷瘤小鼠的肝脏、肾脏和胸腺毒性。
Objective To investigate the effect of Danggui Beimu Kushen pill (angelica, fritillaria, sophorae) on the pathological morphology of tumor, liver, kidney and thymus of cisplatin-treated H22 tumor-bearing mice, and explore its effect-enhancing and toxic-reducing action for cisplatin. Methods H22 tumor-bearing mice models were established, and randomized into model group, cisplatin group, Danggui Beimu Kushen pill group, Danggui Beimu Kushen pill combined cisplatin group, with normal group as control. Drugs were administrated from the eighth day for 14 d, the tumor volume of tumor-bearing mice was dynamically monitored. On the 15th days, the mice were killed to observe the pathological changes of tumor, liver, kidney and thymus under the microscope. Results After treatment for 10 d, compared with model group, the tumor volume of each treatment group was significantly decreased (P〈0.01), Danggui Beimu Kushen pill group was most obvious. Compared with model group, the tumor cellularities, atypia and irregular mitosis of each treatment group were significantly decreased (P〈0.01), the effect of Danggui Beimu Kushen pill combined cisplatin group was better than that of cisplatin group, accompanied by obvious lymphocyte infiltration and hyperplasia of fibrous tissue (P〈0.01). The pathomorphological damages of liver, kidney and thymus in cisplatin group were obviously aggravating than normal group and model group (P〈0.01), however the degree of damages in Danggui Beimu Kushen pill combined cisplatin group were reduced (P〈0.05). Conclusion Danggui Beimu Kushen pill can not only enhance the antitumor effect of cisplatin, but also reduce cisplatin toxicity to liver, kidney and thymus of H22 tumor-bearing mice.
