摘要:目的:寻找适合的治疗海洛因依赖的方法并阐述穴位埋线法控制海洛因大脑海马区稽延性戒断症状的机制。方法:对大鼠采用剂量递增方法皮下注射海洛因建立大鼠海洛因依赖模型,随机分为模型组、自然戒断组、美沙酮治疗组、针刺治疗组和穴位埋线治疗组,并比较各组大鼠戒断症状和信使核糖核酸(Messenger Ribonucleic Acid,mRNA)和蛋白水平表达的情况。结果:我们发现穴位埋线和针刺都和美沙酮一样能有效控制对海洛因依赖大鼠稽延性戒断症状,各个治疗组大鼠海马区的胆囊收缩素(Cholecystokinin,CCK)和C-fos因子在mRNA和蛋白水平表达量都较模型组偏低,说明治疗后大鼠海马区因海洛因依赖受损大脑功能正在逐步恢复,证明我们的治疗是有效的。结论:穴位埋线能控制大鼠海洛因依赖稽延性戒断症状,同时,可以降低大鼠海马区神经元CCK和C-fos表达水平来修复大脑由于海洛因依赖所受损伤。
Objective:To find appropriate treatment for heroin dependence and illustrate the mechanism of acupoint embedding for the control of delayed withdrawal symptoms in the hippocampus of heroin in brain.Methods:We set up a model of rat heroin dependence by incremental injection of heroin subcutaneous.Then we randomly divided the rats into the model group,the natural withdrawal group,the methadone treatment group,the acupuncture group and the acupoint embedding group.The withdrawal symptoms and the expression of mRNA and protein levels in each group were compared.Results:We found out that just as the methadone and the acupuncture the acupoint embedding can control prolonged withdrawal symptoms in heroin dependent rats effectively.After treatment the expression of CCK and C-fos in treatment groups were lower than that in the model group.It showed that the brain function in rats with impaired heroin dependence was gradually recovering after treatments.These results suggest that our treatments are effective.Conclusion:Acupoint embedding can control prolonged withdrawal symptoms in heroin dependent rats and the therapy also can reduce of neuronal CCK and C-fos expression levels in hippocampus to repair brain damage from heroin dependence.
