摘要:目的:观察关节内注射川芎嗪对鼠骨关节炎(Osteoarthritis,OA)模型关节软骨组织病理学及白细胞介素-1β(IL-1β)表达的影响,探讨川芎嗪对OA的修复作用。方法:50只SD大鼠随机分为空白组、模型组、透明质酸钠组、丹参组、川芎嗪组5组,每组10只,除空白组外,其余各组作左侧跟腱切除建立右膝OA模型。造模后4周,每组随机处死动物2只,以检测动物模型是否成功;剩余40只于造模后第5周开始,空白组、模型组右膝关节内注射生理盐水,透明质酸钠组、丹参组、川芎嗪组右膝关节内分别注射相应药物。于第12周末全部处死动物,取股骨外髁软骨标本,进行苏木精-伊红、Masson染色,光镜下观察其病理改变,以免疫组织化学法检测关节软骨中的IL-1β水平。结果:与模型组比较,川芎嗪组关节软骨破坏减轻,Ⅱ型胶原脱失轻微,Mankin’s评分明显改善(P<0.01),IL-1β的表达水平显著降低(P<0.01)。结论:川芎嗪能明显降低OA软骨退变的程度,阻止软骨中正常Ⅱ型胶原向Ⅰ型胶原的转变,明显降低OA软骨中IL-1β的水平,具有保护和修复早期OA关节软骨的作用。
Objective:To observe the effect of Ligustrazine on histopathology and expression of interleukin-1β(IL-1β) in cartilage in rat model of OA;and to observe the repair of Osteoarthritis(OA) by injecting Ligustrazine into the joint cavity.Methods:The model of right knee OA was made by the way of cutting off the left achilles's tendon.Animals in Ligustrazine group were injected with Ligustrazine hydrochloride.At the end of the experiment,HE and Masson staining of the articular cartilage were made to observe pathology changes under a light microscope and the distribution of IL-1β in cartilage by the immunohistochemistry.Then Mankin's score and the level of IL-1β of cartilage were performed.Results:The destruction of cartilage was alleviated and the loss of typeⅡcollagen was depressed in Ligustrazine group.The Mankin's score and the expression level of IL-1β in the cartilage of the Ligustrazine group were significantly decreased(P0.01).Conclusion:Intra-articular injection of Ligustrazine can significantly reduce the severity of cartilage degradation in rat model of OA and prevent the change of collagen from type Ⅱ into type Ⅰ.It can also significantly reduce the levels of IL-1β of cartilage in rat model of OA.It has a protective effect on articular cartilage in rat model of OA at early stage.
