摘要:目的:研究补脑软胶囊对Aβ_(1-42)诱导SH-SY5Y神经细胞损伤的保护作用。方法:建立β-淀粉样蛋白(Aβ_(1-42))诱导SH-SY5Y细胞损伤模型,采用PBS溶解的补脑软胶囊内容物干预24h,采用CCK-8法测定细胞存活率,在Hochest33258染色荧光显微镜下观察SH-SY5Y细胞凋亡情况。结果:SHSY5Y细胞经Aβ_(1-42)诱导损伤后采用补脑软胶囊干预24h,与模型组相比,用药组细胞突触结构消失减少,贴壁状态明显改善,提高了细胞存活率;Hoechst33258染色结果显示,对照组细胞呈现弥散均匀的弱荧光,而Aβ_(1-42)模型组细胞核出现固缩形态和颗粒状荧光,补脑软胶囊干预后可明显减少细胞颗粒状荧光及固缩形态,提示补脑软胶囊可有效抑制Aβ_(1-42)引起的细胞凋亡。结论:补脑软胶囊对Aβ_(1-42)诱导的SH-SY5Y细胞损伤具有一定的保护作用。
Objective:To study the protective effects of Bunao soft capsule on SH-SY5 Ycell injury induced by Aβ_(1-42).Methods:SH-SY5 Ycell models induced by Aβ_(1-42)were established.The contents of Bunaosoft capsule dissolved by PBS were used to intervene for 24 hours.Cell survival rate was measured by CCK-8 method.The nuclear morphology of SH-SY5 Ycells was observed by Hochest33258 staining fluorescence microscope.The protective effect of Bunaosoft capsule on injured SH-SY5 Ycells was analyzed.Results:SH-SY5 Ycells were injured by Aβ_(1-42)and incubated with Bunao soft capsule for 24 hours.Compared with the model group,synaptic structure was clear,cell adherence was improved,and the protective effect of Bunaosoft capsule on nerve cells was accurate.The results of Hoechst33258 staining showed that diffuse and uniform low-intensity fluorescence appeared in the nucleus of normal control group,and the nucleus of model group Aβ_(1-42)showed dense and dense solid-shrinkage morphology and granular fluorescence.Bunao soft capsule could significantly reduce the solid-shrinkage morphology and granular fluorescence of SH-SY5 Ycells after intervention.Bunaosoft capsule could effectively inhibit the apoptosis induced by Aβ_(1-42).Conclusion:Bunaosoft capsule has obvious protective effect on SH-SY5 Ycell injury induced by Aβ_(1-42).
