摘要:目的观察固本增骨方对去卵巢大鼠血清BGP、TRACP-5b及骨质量的影响,探讨固本增骨方防治绝经后骨质疏松症的作用机制。方法卵巢切除法建立大鼠骨质疏松模型,随机分为模型对照组、戊酸雌二醇组、固本增骨方高、中、低浓度组,选取同龄大鼠作为空白对照组。用药干预12周后,腹腔麻醉,采用双能X线骨密度测量仪计算大鼠全身骨密度;ELISA法检测大鼠血清骨生成代谢指标BGP及骨吸收代谢指标TRACP-5b;解剖分离左侧股骨标本,置于AG-X系列台式电子万能试验机,计算骨生物力学性能指标;激光共聚焦显微镜扫描各组股骨组织游离[Ca^[2+]]i浓度。结果与空白对照组相比,模型组大鼠全身骨密度值显著下降,股骨最大载荷和弹性模量明显降低,差异有统计学意义(P<0.05)。与空白对照组比较,模型组大鼠BGP、TRAP-5b含量明显升高(P<0.05);给予药物干预后,与模型组相比,戊酸雌二醇组和固本增骨方各组BGP、TRAP-5b含量均明显下降(P<0.05);且固本增骨方高浓度组优于低浓度组(P<0.05);与模型组对比,戊酸雌二醇组和固本增骨方不同浓度组可显著增加全身骨密度值,明显提高股骨最大载荷和弹性模量、[Ca^[2+]]i浓度,差异有统计学意义(P<0.05)。固本增骨方高浓度组与低浓度组比较有统计学差异(P<0.05),呈量效依赖关系。固本增骨方各组与戊酸雌二醇组比较差异无统计学意义(P>0.05)。结论固本增骨方可降低去卵巢大鼠模型血清BGP、TRAP-5b含量,改善全身骨密度,提升骨生物力学性能,增加骨组织中游离[Ca^[2+]]i浓度,从而发挥对绝经后骨质疏松症的防治作用。
Objective To observe the effect of Guben Zenggu Decoction on serum BGP, TRACP-5 b and bone quality in ovariectomized rats, and to explore the mechanism of Guben Zenggu Decoction in preventing and treating postmenopausal osteoporosis. Methods The osteoporosis model was established by ovariectomy. The models were randomly divided into model control group, estradiol valerate group, and groups with high, medium and low concentrations of Guben Zenggu Decoction. Selected the same age Rats as the blank control group. After the intervention for 12 weeks, abdominal anesthesia was performed, bone mineral density(BMD) of rats was measured in vivo by dual-energy X-ray bone mineral density(DXBMD) measuring instrument. BGP and TRACP-5 B of bone resorption and metabolism were detected by ELISA. Left femoral specimens were dissected and placed on AG-X series desktop electronic universal testing machine to calculate the biomechanical properties of bone. Free [Ca^[2+]]i concentration of femoral tissue was scanned by confocal laser microscopy in each group. Results Compared with the blank control group, the bone mineral density(BMD) and the maximum load and modulus of elasticity of femur in the model control group decreased significantly(P< 0.05).Compared with the blank control group, the content of BGP and TRAP-5 b in the model control group increased significantly(P< 0.05). After drug intervention, the content of BGP and TRAP-5 b in estradiol valerate group and Guben Zenggu Decoction groups decreased significantly compared with the model control group(P<0.05). Moreover, the high concentration group of Guben Zenggu Decoction was superior to the low concentration group(P< 0.05). Compared with model control group, estradiol valerate group and Guben Zenggu Decoction with different concentration groups could significantly increase the whole body BMD value, significantly increase the maximum load, elastic modulus of femur and[Ca^[2+]]i concentration, with statistically significant differences(P< 0.05). There were statistically significant differences between the high concentration group and the low concentration group of Guben Zenggu Decoction(P< 0.05), showing a dose-effect dependent. There was no significant difference between Guben Zenggu Decoction groups and estradiol valerate group(P >0.05). Conclusion Guben Zenggu Decoction can reduce serum BGP and TRAP-5 b content in ovariectomized rat model, improve the whole body bone mineral density of ovariectomized rats, effectively enhance the biomechanical properties of bone, Increasing free [Ca^[2+]]i concentration in bone tissue, so as to play a role in preventing and treating postmenopausal osteoporosis.
