摘要：铁死亡是一种铁过载诱导的细胞膜脂质过氧化物蓄积驱动的细胞死亡模式,参与疾病的发生发展过程.研究显示,铁死亡在帕金森病(Parkinson′s disease,PD)过程中起着关键作用,中医药在PD的治疗方面具有独特优势,以铁死亡理论为指导的中医药治疗PD是当今研究的热点内容.本文作者阐述了铁死亡的调控机制,与PD的关系,探讨了中医药基于铁死亡理论治疗PD的研究进展.大量研究表明,铁死亡的发生与铁代谢失衡、脂质过氧化、氨基酸代谢异常等途径有关,铁死亡在PD的发展和治疗过程中均展现出重要的调控作用.研究发现,中药有效成分、中药复方和针灸通过多靶点、多途径抑制铁死亡治疗PD,为PD的研究提供科学依据和参考.目前,中医药调控铁死亡治疗PD具有良好的研究前景,但要实现临床转化,仍面临着许多挑战.
Ferroptosis is a form of cell death driven by iron overload-induced accumulation of lipid peroxides on cell membranes,playing a role in the onset and progression of diseases.Studies indicate that ferroptosis plays a critical role in Parkinson′s disease(PD),and traditional Chinese medicine(TCM)offers unique advantages in PD treatment.TCM-based interventions targeting ferroptosis have become a key research focus in PD management.This study elaborates on the regulatory mechanisms of ferroptosis,its association with PD,and recent advances in TCM strategies against PD guided by ferroptosis theory.Many researches suggest that ferroptosis is linked to dysregulated iron metabolism,lipid peroxidation,and abnormal amino acid metabolism,demonstrating significant regulatory effects in both PD pathogenesis and treatment.Research has revealed that active components of Chinese herbs,herbal formulations,and acupuncture can inhibit ferroptosis through multiple targets and pathways,providing a scientific basis for PD therapy.Currently,TCM-mediated regulation of ferroptosis in PD treatment shows promising potential,yet clinical translation still faces considerable challenges.
