摘要:目的:观察黄芩素对模型大鼠耳廓复合痤疮的影响,探究黄芩素对痤疮的干预机制。方法:按随机数字表法从64只雌雄各半的Wistar大鼠中抽取10只作为空白组,对其余54只大鼠用kligman法建立右耳廓复合痤疮模型,21 d后随机抽取4只进行HE观察,确认造模成功后随机分为模型组、黄芩素高、中、低剂量组以及阳性对照组各10只。低、中、高剂量组分别给予不同浓度的黄芩素,阳性对照组给予0.025%的维A乳酸膏,均每日给药1次,连续用药4周,其余2组不给予药物治疗,4周后对大鼠进行局部组织采样,并采用ELISA法检测样品组织中IL-4以及MMP-9的含量。结果:模型组IL-4、MMP-9的含量与空白组比较差异显著(P<0.01),黄芩素低、中、高剂量组及阳性对照组大鼠组织中IL-4、MMP-9的含量较模型组均有不同程度的降低(P<0.05),各组含量趋势为高剂量组<阳性对照组<中剂量组<低剂量组。结论:黄芩素治疗痤疮疗效确切,可能与黄芩素降低局部组织中IL-4、MMP-9有关。
Objective:To observe the effects of baicalein on rat auricle complicated acne model and to investigate interventional mechanism of baicalein on acne model.Methods:Random number table was used to select 10 of the 64 male and female rats as blank group,and the auricles acne of the remaining 54 rats were modeled by kligman method.After 21 days,4 rats were randomly selected for observation by Hematoxylin-Eosin stain(HE).After confirming the success of modeling,they were randomly divided into model group,baicalein high,medium,low-dose group and positive control group,10 rats in each group.Low-dose groups,medium-dose groups,and high-dose groups were given baicalein of different concentrations,and the positive control group was given 0.025%tretinoin cream,all of which were given once a day for four consecutive weeks.The remaining two groups were not given drug treatment.The rats were subjected to local tissue sampling,and the content of IL-4 and MMP-9 in the sample tissues were detected by ELISA.Results:The levels of IL-4 and MMP-9 in the model group were significantly different from those in the blank group(P<0.01).Compared with the model group,the levels of IL-4 and MMP-9 in the rat tissues of baicalein low,medium,high-dose groups and the positive control group were decreased significantly(P<0.05),and the trend of each group was high-dose group
