摘要:目的:探讨红芪多糖在2型糖尿病胰岛素抵抗中的防治效果及作用机制。方法:取50只wistar大鼠,随机选取10只作为空白对照,其余40只一次性注射链脲佐菌素(STZ)30mg/kg,并饲以高脂高糖饲料6周制作2型糖尿病胰岛素抵抗模型。6周后选取空腹血糖≥11.1mmol/L的大鼠随机分为模型组、红芪多糖组、二甲双胍组。各组分别灌服生理盐水、红芪多糖、二甲双胍1月。1月后取血清测空腹胰岛素、空腹血糖、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)的含量,并计算胰岛素敏感指数(ISI)。结果:与模型组相比,红芪多糖能显著降低2型糖尿病胰岛素抵抗大鼠空腹血糖(P<0.05)。红芪多糖能调节2型糖尿病胰岛素抵抗大鼠的血脂紊乱,降低血清总胆固醇(P<0.05)和甘油三酯(P<0.05),提高了高密度脂蛋白的含量(P<0.05),提高了胰岛素的敏感指数。结论:红芪多糖对2型糖尿病胰岛素抵抗大鼠有降低血糖、调节血脂紊乱的作用,并改善了胰岛素敏感指数,从而对2型糖尿病胰岛素抵抗具有一定的防治作用。
Objective: To investigate the effects of Radixhedysari polysaccharide(RP) on insulin sensitivity in T2DM rats with insulin resistance and the primary pharmacological mechanism.Methods: Ten rats were selected randomly as free group from fifty rats accord to sex and weight,others which make the model for type 2 diabetic insulin resistance(T2D-IR) rats were performed through intraperitonea injection of lower dosage of streptozocin(STZ) 30mg/kg and high caloric diet(abounded with sugar fat)six weeks.Then the rats whose free blood glucose(FBG) equals or surpasses 11.1mmol/L were selected and divided into model group,RP group,Metformin(MT)group randomly accord to sex and weight.After respectly giving fresh water,RP,MT for 30 days,the effects of RP on blood and/or serum contents of glucose、lipids、insulin and insulin sensitive index(ISI) were observed in the model rats.Results:RP insignificantly fasting free blood glucose(FBG);decreased markedly the levels of total cholesterols(TC) and triglyceride(TG),and increase the content of high-density lipoprotein cholesterol(HDL-C);and reduced the concentration of insulin in hyperinsulinemia,and potentiated ISI in T2D-IR.Conclusion:Effects in the experiment with type 2 diabetic-resistant rats,RP can reduce level of blood glucose,regulate components and content of blood lipids,and improve insulin sensitity.
