摘要:目的:优选荆芥药材挥发油的提取工艺,分析其化学成分,并对其含有的主要化学成分胡薄荷酮进行含量测定。方法:采用酶辅助水蒸气蒸馏法提取荆芥药材挥发油,结合Box-Behnken响应面法优化提取工艺;通过气相色谱-质谱法(GC-MS)分析水蒸气蒸馏法、酶辅助水蒸气蒸馏法提取的荆芥药材挥发油的化学成分,并利用高效液相色谱法(HPLC)测定2种不同提取方法所得荆芥药材挥发油中胡薄荷酮的含量。结果:最佳提取工艺为纤维素酶添加量1.563%、酶解时间0.8 h、酶解温度44℃,挥发油平均得率为0.779%,与传统水蒸气蒸馏法(0.540%)相比挥发油平均得率增加较多。共鉴定出荆芥药材挥发油中26个化学成分,主要化学成分为L-薄荷酮(43.60%)和胡薄荷酮(36.91%)。方法学考察结果显示,胡薄荷酮质量浓度为0.0472~5.6670 mg·mL^(–1)时,线性关系良好(r=0.9998),精密度(RSD=1.01%)、重复性(RSD=1.14%)及回收率(96.38%~103.55%,RSD=3.88%)均符合要求。结论:酶辅助水蒸气蒸馏法提取工艺可有效提高荆芥药材挥发油得率,且对主要成分相对质量分数影响较小;建立的基于HPLC的含量测定方法准确可靠,适用于对荆芥药材挥发油中胡薄荷酮进行含量测定。
Objective:To optimize the extraction process,analyze the chemical composition,and determine the major component content of Schizonepetae Herba essential oil(STEO).Methods:The cellulase-assisted steam distillation method was utilized for STEO extraction,with process parameters optimized using Box-Behnken response surface methodology.The chemical components of the volatile oil samples obtained from Schizonepetae Herba by steam distillation and enzyme-assisted steam distillation were analyzed by gas chromatography-mass spectrometry(GC-MS).Additionally,the content of pulegone in the volatile oil samples derived from these two extraction methods was determined by highperformance liquid chromatography(HPLC).Results:The optimal extraction conditions were determined as follows:enzyme concentration of 1.563%,enzymatic hydrolysis time of 0.8 h,and reaction temperature of 44℃.Under these conditions,the average extraction yield of STEO was 0.779%,which was significantly higher than that of conventional steam distillation(0.540%).GC-MS revealed 26 chemical components in STEO,with L-menthone(43.60%)and pulegone(36.91%)being the predominant compounds.HPLC validation demonstrated excellent linearity(r=0.9998)for pulegone within the concentration range of 0.0472-5.6670 mg·mL^(–1).The precision(relative standard deviation,RSD=1.01%),repeatability(RSD=1.14%),and recovery rates(96.38%-103.55%,RSD=3.88%)all met the requirements specified in pharmacopeial standards.Conclusion:The cellulase-assisted extraction process can effectively increase the yield of volatile oils from Schizonepetae Herba,with no significant impact on the relative content of its main components.The established HPLC method is accurate and reliable for the quantification of pulegone,providing a robust analytical framework for the quantitation of pulegone in STEO.
