摘要:目的:探讨附子与瓜蒌不同配比对小鼠急性毒性的影响。方法:将生附子、全瓜蒌粉碎成40目细粉,取不同配比(1∶0、2∶1、1∶1、1∶2、0∶1)的附子、瓜蒌,加相当于药材总质量的16倍水,浸泡1小时,置95℃水浴锅中,待药液温度上升至75℃时开始计时,水浴加热10分钟,离心(3 000 r/mi n)10分钟,取上清液,低温减压浓缩,将其配成所需浓度的药液给小鼠灌胃,运用"改进寇氏法"检测各组药液的LD50。结果:1∶0、2∶1、1∶1、1∶2组LD50分别为(4.50±0.89)(4.72±0.79)(4.98±0.76)(4.63±0.65)(含附子)g/kg,全瓜蒌组无死亡,未测出LD50。结论:附子与瓜蒌不同配比(1∶0、2∶1、1∶1、1∶2)小鼠LD50无明显区别,提示附子与瓜蒌配伍有可能不发生"相反"作用。
Objective: To explore the effects of FuZi (Radix Aconiti Lateralis Preparata) and GuaLou (Trichosanthes kirilowii Maxim) in different proportions on acute toxicity of mice. Method: FuZi and GuaLou were powdered into the granule of 380μ m and chosen in different proportions (1:0, 2:1, 1:1, 1:2, 0:1), water was added equaled to 16 times of total quality of drugs, the powder was dipped in water for one hour and boiled in the water bath at 75℃, when the temperature of decoction rose to 75℃, the time was counted down. The water-bath washeated for ten minutes, centrifugation for ten minutes (3 000 r/min). Liquid supematant was got and abstracted in the low temperature and pressure, given intragastrically to the mice. LD50 of the liquid in different groups were detected with acute toxicity testing. Result: LD50 of 1 :0, 2:1, 1:1 and 1:2 groups were respectively(4.50±0.89),(4.72±0.79),(4.98±0.76) (4.63±0.65) ,(including FuZi)g/kg, there was no death in GuaLou group and LD50 was not detected out. Conclusion: LD50 of mice in different proportions (1:0, 2:1, 1:1, 1:2) of FuZi and GuaLou shows no significant difference, which suggests that the compatibility of FuZi and GuaLou might be not opposite.
